快速心房起搏对家兔心房Akt和GSK-3β活性的影响
Effects of rapid atrial pacing on the activities of atrial Akt and GSK-3β in rabbits
【摘要】 [目的]探讨快速心房起搏对家兔心房Akt和GSK-3β活性的影响.[方法]经右颈外静脉插入电极至右心房,制作快速心房起搏家兔房颤模型,观察心房快速起搏8 h后心房组织形态学、Akt和GSK-3β活性变化情况.[结果]快速起搏8 h组家兔血清中ANP水平较假手术组显著升高(P<0.05);HE染色观察结果显示,快速起搏8 h组心房心肌纤维排列紊乱,间质水肿;透射电子显微镜观察结果显示,心房肌线粒体嵴消失、变形、空泡化形成,肌浆网扩张,糖原颗粒聚集明显;Western blot检测结果显示,快速起搏8 h组心房组织Akt和GSK-3β表达水平较假手术组显著降低(P<0.05).[结论]快速心房起搏诱导的Akt和GSK-3β活性降低与房颤心房结构重构有关系.
【Abstract】 OBJECTIVE To explore the effects of rapid atrial pacing on the activities of atrial Akt and GSK-3β in rabbits. METHODS The atrial fibrillation model of rabbits with rapid atrial pacing was established by inserting electrodes into the right atrium via the right external jugular vein. Atrial histomorphology, the changes of Akt and GSK-3β activities were observed after 8 h of rapid atrial pacing. RESULTS The serum ANP level was significantly higher in the rapid atrial pacing 8 h group than that in the sham operation group(P<0.05). The results of HE staining showed that atrial myocardial fibers were disordered and interstitial edema occurred in the rapid atrial pacing 8 h group. The results of transmission electron microscope showed that the mitochondrial ridge of atrial muscle disappeared, deformed and vacuolated, the sarcoplasmic reticulum expanded, and the glycogen granules accumulated obviously. Western blot results showed that the expression levels of Akt and GSK-3β in the atrial tissue of the rapid atrial pacing 8 h group were significantly lower than those of the sham operation group(P<0.05). CONCLUSION Decreased Akt and GSK-3β activity induced by rapid atrial pacing was associated with atrial structural remodeling in atrial fibrillation.
