【摘要】 目的观察人肺癌A549细胞形态,探讨黄芪注射液对人肺癌A549细胞形态和增殖的影响。方法以体外培养的人肺癌A549细胞为研究对象,倒置显微镜观察细胞形态;采用活细胞计数试剂盒(CCK-8)法检测黄芪注射液对A549细胞增殖的影响;采用改良寇氏法计算黄芪注射液作用A549细胞的半数抑制浓度(IC50)。结果黄芪注射液作用A549细胞24、48 h时,100、200、400 mg/mL黄芪注射液组对其均有促生长的作用,但随着浓度的增加而减弱,600、800 mg/mL黄芪注射液组对细胞有抑制作用;作用72 h后,400 mg/mL黄芪注射液组不再具有促细胞生长作用,600、800 mg/mL黄芪注射液组的抑制作用增强。改良寇氏法计算24、48、72 h的IC50分别为776、851、616 mg/mL。结论低浓度的黄芪注射液在短时间内对A549细胞有促生长的作用,达到一定作用时间后,浓度越高对A549细胞的抑制作用越明显。更多还原

【Abstract】 Objective To observe the morphological changes of human lung cancer A549 cells and to investigate theeffect of astragalus injection on the morphology and proliferation of human lung cancer A549 cells. Methods Human lung can-cer A549 cells were cultured in vitro,the morphology of A549 cells was observed by inverted microscope;the effect of astraga-lus injection on the proliferation of A549 cells was detected by CCK-8 method;the modified Kou′s method was used to calcu-late the median-inhibitory concentration(IC50)to research the effect of astragalus injection on A549 cells. Results The effectof astragalus injection on A549 cells at 24,48 h by the mass concentration of 100,200 mg/mL and 400 mg/mL astragalus injec-tion could promote cells growth,but as the concentration increased,the promotion decreased,the mass concentration of 600,800 mg/mL astragalus injection had inhibitory action on cells;when the reaction time of 72 h,the mass concentration of 400 mg/mLastragalus injection had no a growth promoting effect on cells;and the inhibitory action on cells of the mass concentration of 600,800 mg/mL astragalus injection were enhanced. The IC50 of 24,48,72 h were 776,851,616 mg/mL respectively. Conclusion Low concentration of astragalus injection can promote the growth of A549 cells in a short time. After a certain actiontime,the higher the concentration,the greater the inhibition effect of A549 cells.更多还原
