IL-10基因的单核苷酸多态性与儿童哮喘的相关性研究
Study on the correlation between single nucleotide polymorphisms of IL-10 gene and childhood asthma
【摘要】 目的探讨白细胞介素-10(IL-10)基因单核苷酸多态性(SNP)与儿童哮喘的关联性。方法选取该院2004年1月至2005年8月收治的84例儿童哮喘患儿为哮喘组,选取同期116例无血缘关系的健康体检儿童作为对照组。采用基于基质辅助激光解吸电离-飞行时间质谱技术检测IL-10基因SNP(SNP位点包括rs1800871、rs1800872、rs1800896、rs3024492和rs3024496)的分型变化,采用免疫发光法测定血清总Ig E水平,以Ig E水平超过1 000 k U/L为高Ig E水平。结果 SNP基因频率达到Hardy-Weinberg遗传平衡(P>0.05),哮喘组rs1800871、rs1800872、rs1800896、rs3024492和rs3024496这5个位点的基因型构成比与对照组比较,差异无统计学意义(P>0.05)。rs1800871位点的TT基因型携带者和rs1800872的AA基因型携带者更容易表现为高Ig E水平(OR=5.000、4.599,95%CI 1.926~24.709、1.012~22.452,P=0.033、0.046)。结论 IL-10基因的SNP与儿童哮喘的发病不相关,但与哮喘儿童的血清高Ig E水平相关。rs1800871位点的TT基因型和rs1800872位点的AA基因型可能是哮喘儿童高Ig E水平的易感基因型。
【Abstract】 Objective To investigate the correlation between gene single nucleotide polymorphism(SNP)of interleu-kin-10(IL-10)and childhood asthma. Methods A total of 84 cases childhood asthma were selected as asthma group and 116 cases unrelated healthy children as control group from January 2004 to August 2005 in People′ s Hospital of Guangxi Zhuang Autonomous Region. The typing changes of SNP(the locus of SNP were rs1800871,rs1800872,rs1800896,rs3024492 andrs3024496)in IL-10 gene were detected by matrix assisted laser desorption ionization time-of-flight mass spectrometry,serumtotal Ig E level in childhood asthma was measured by immunity the luminescence method,the Ig E level more than 1 000 k U/Lwas high Ig E level. Results The genotypic distributions of SNP was statistical according to Hardy-Weinberg equilibrium(P>0.05),the ratio of genotype in the 5 locuses(rs1800871,rs1800872,rs1800896,rs3024492 and rs3024496)in the asthmagroup compard with those in control group,the differences were not statistically significant(P>0.05). The TT genotype at thers1800871 locus carriers and the AA genotype at the rs1800872 locus carriers were more likely to display high Ig E level(OR=5.000,4.599;95%CI 1.926~24.709,1.012~22.452;P=0.033,0.046). Conclusions SNP in IL-10 gene is not related to child-hood asthma,but is related to serum high Ig E level in childhood asthma. The TT genotype at the rs1800871 locus and the AAgenotype at the rs1800872 locus may be susceptible genotypes of high Ig E level in childhood asthma.
