肺气虚寒型变应性鼻炎患者Thl7/Treg细胞中转录因子表达的差异性研究

【摘要】 目的探讨Thl7细胞中转录因子(ROR-γt)及Treg细胞中转录因子(Foxp3)在不同分类与分度的肺气虚寒型AR患者外周血中的表达。方法选取未经治疗的肺气虚寒型AR患者19例,健康志愿者10例作对照,肺气虚寒型AR患者组进行分类及分度为四组:轻度间歇性、中-重度间歇性、轻度持续性及中-重度持续性,实时荧光定量PCR(RT-qPCR)法检测各组外周血Thl7细胞及Treg细胞中转录因子ROR-γt mRNA、Foxp3 mRNA的表达。结果 19例肺气虚寒型AR患者外周血中,ROR-γt mRNA表达明显高于对照组(*P<0.05),而Foxp3 mRNA表达显著低于对照组(**P<0.01),ROR-γt/Foxp3 mRNA水平显著高于对照组(**P<0.01)。不同分类与分度的肺气虚寒型AR患者,中-重度持续性AR组ROR-γt mRNA、ROR-γt/Foxp3mRNA表达显著高于对照组(**P<0.01),而Foxp3 mRNA表达明显低于对照组(*P<0.05),中-重度持续性AR组ROR-γt mRNA表达显著高于轻度间歇性、中-重度间歇性及轻度持续性(**P<0.01),中-重度持续性AR组Foxp3 mRNA表达显著低于轻度间歇性(**P<0.01),中-重度持续性AR组ROR-γt/Foxp3 mRNA表达显著高于轻度间歇性、中-重度间歇性及轻度持续性(**P<0.01)。结论肺气虚寒型AR患者Th17/Treg细胞中转录因子ROR-γt/Foxp3 mRNA表达失衡,中-重度持续性的肺气虚寒型AR患者失衡显著,但轻度间歇性、持续性、中-重度间歇性的肺气虚寒型AR患者ROR-γt/Foxp3 mRNA表达失衡不显著。

【Abstract】 Objective To investigate the expression of transcription factor(ROR-γt) in Thl7 cells and transcription factor(Foxp3) in Treg cells in peripheral blood of patients with allergic rhinitis of lung qi deficiency and cold type with different classification and degree. Methods The selection of the treatment of 19 cases of patients with allergic rhinitis of lung qi deficiency and cold, 10 healthy volunteers as control group. The patients with allergic rhinitis type classification and dividing into 4 groups: Mild intermittent allergic rhinitis、Moderate to severe intermittent allergic rhinitis、Mild persistent allergic rhinitis and Moderate to severe persistent allergic rhinitis, RT-qPCR method was used to detect the expression of the peripheral group blood Thl7 and Treg cells of transcription factor ROR-γt mRNA and Foxp3 mRNA. Results Nineteen cases of lung qi deficiency and cold in the peripheral blood of allergic rhinitis patients, the expression of ROR-γt mRNA was significantly higher than the control group(*P<0.05), and Foxp3 mRNA expression was significantly lower than the control group(**P<0.01), ROR-γt/Foxp3 mRNA level was significantly higher than the control group(**P<0.01). Different classifications and grades of patients with allergic rhinitis of lung qi deficiency and cold, moderate to severe persistent expression of allergic rhinitis group ROR-γt mRNA, ROR-γt/Foxp3 mRNA was significantly higher than the control group(**P<0.01), and Foxp3 mRNA expression was significantly lower than the control group(*P<0.05), moderate to severe persistent allergic rhinitis group expression of ROR-γt mRNA was significantly higher than mild intermittent and moderate to severe intermittent and mild persistent(**P<0.01), moderate to severe persistent expression of allergic rhinitis group Foxp3 mRNA was significantly lower than mild intermittent group(**P<0.01), moderate to severe persistent allergic rhinitis group expression of allergic rhinitis group ROR-γt/Foxp3 mRNA were significantly higher than that of mild intermittent and moderate to severe intermittent and mild persistent(**P<0.01). Conclusion There is a imbalance of the transcription factor ROR-γt/Foxp3 mRNA expression of Th17/Treg cells in patients with allergic rhinitis of lung qi deficiency and cold. Significant imbalance is found in moderate to severe of lung qi deficiency and cold type allergic rhinitis patients, but not in mild intermittent and persistent and moderate to severe intermittent cases.
