紫外线和热力对55型腺病毒的灭活效果
Inactivation effect of UV and heat on adenovirus 55
【摘要】 目的探讨紫外线和热力对55型腺病毒(HAdV-55)的灭活效果,为HAdV-55感染的防控提供参考。方法分别将HAdV-55置于波长245 nm的紫外线和65℃热力条件下作用30 min、1 h和2 h,通过定量PCR和病毒滴度试验,检测各处理因素对HAdV-55感染HEp-2细胞水平的影响。结果紫外线照射30 min后,HAdV-55感染HEp-2细胞的水平降低约50%;紫外照射1 h和2 h后,病毒仍具有一定感染能力,但感染水平显著降低。65℃热力处理30 min,HAdV-55感染HEp-2的水平降低了约104个数量级。紫外线和热力灭活的病毒产生子代病毒的能力均显著降低。结论紫外线和热力对HAdV-55都有灭活作用,其灭活作用的强弱与处理时间有关,254 nm紫外线照射和65℃热力处理30 min以上均能显著降低HAdV-55产生子代病毒的水平。
【Abstract】 Objective To explore the inactivation effects of UV and heat on human Adenovirus 55(HAdV-55) and provide references for prevention and control of HAdV-55 infection. Methods HAdV-55 was placed under 245 nm UV and 65℃ heat for 30 min, 1 h and 2 h, respectively. Quantitative PCR and virus titer test were performed to examine the impacts of various factors on HAdV-55-infected HEp-2 cells. Results After 30 min’s UV action, the level of HAdV-55-infected HEp-2 cells decreased by 50%approximately; after 1 h and 2 h’s UV action, the virus was still infectious to some extent, but the level of infection decreased greatly.After 30 min’s heat treatment at 65 ℃, the level of HAdV-55-infected HEp-2 cells decreased by about 104 orders of magnitude.Virus inactivated by UV and heat showed a significantly lowered capability of generating progeny virus. Conclusion Both UV and Heat can inactivate HAdV-55. The effect of inactivation depends on the time of action. More than 30 min’s 254 nm UV action and heat treatment at 65℃ can considerably reduce the capability of HAdV-55 to generate progeny virus.
