多重PCR方法检测沙门氏菌血清型适用性探讨
Applicability of multiple PCR assay for Salmonella enteric serotyping
【摘要】 目的探讨在食源性疾病病例监测中,多重PCR方法用于沙门氏菌的血清型分型的适用性。方法利用多重PCR方法对113株分离自食源性疾病病例的沙门氏菌进行血清分型,得到菌株可能血清型。同时,菌株血清型经Luminex x MAP()沙门氏菌血清分型法及传统的血清玻片凝集方法最终确定。比较可能血清型与菌株确定血清型间的符合程度,探讨多重PCR方法用于沙门氏菌血清分型的适用性。结果在113株菌株中共检出23个沙门血清型。多重PCR方法可准确推测其中11个血清型,涵盖了113株中的94株(83. 2%)。结论多重PCR方法为食源性疾病监测中的沙门氏菌血清分型提供了简便快速、价格低廉的辅助方式,利于在基层公共卫生实验室推广。
【Abstract】 Objective To investigate the applicability of multiple PCR assay to serotyping Salmonella enterica in the patients who suffering from foodborne disease in central China. Methods A total of 113 strains of Salmonella enterica isolated from foodborne disease patients collected through CFSA( China National Center for Food Safety Risk Assessment) foodborne disease surveillance system were included in the study. The possible serotypes of those strains were analyzed with multiple PCR assay.At the same time,the strain serotypes were finally confirmed using the Luminex x MAP( ) Salmonella Serotyping Assay( x MAP SSA) and serum slide agglutination test. To explore the applicability of the multiple PCR assay,the consistency between possible serotypes and finally confirmed serotypes was compared. Results The 113 samples contained 23 serotypes.11 serotypes,which covered 94 of all 113 strains( 83. 2%),were accurately identified using the multiple PCR assay.Conclusion This study shows that the multiple PCR assay provides a time-and cost-effective auxiliary means for serotyping Salmonella enterica from foodborne disease,particularly,in the grass-roots regionalism.
