超高效液相色谱-四极杆飞行时间串联质谱法鉴定α-鹅膏毒肽在大鼠体内的代谢产物
Identification of metabolites of α-amanitin in rat urine by ultra performance liquid chromatography-electrospray ionization quadrupole time-of-flight mass spectrometry
【摘要】 目的检测和鉴定大鼠尿中α-鹅膏毒肽的主要代谢产物,探讨α-鹅膏毒肽在大鼠体内的主要代谢途径。方法大鼠以0. 5 mg/kg的剂量口饲α-鹅膏毒肽,分别采集实验组与对照组的尿液样品,尿液样品经简单处理后采用超高效液相色谱-四极杆飞行时间串联质谱仪检测,利用碰撞能量梯度(MSE)、质量亏损过滤(MDF)和离子色谱峰提取等技术处理数据。结果在大鼠尿中共检测到8种代谢产物,其中,Ⅰ相代谢产物5种,Ⅱ相代谢产物3种,同时鉴定了代谢产物的结构,推测α-鹅膏毒肽在大鼠体内可能的主要代谢途径有氧化、脱氢、脱羧、甲基化反应。尿中主要代谢产物为原形、氧化代谢产物、脱氢代谢产物、脱羧代谢产物、甲基化结合物,α-鹅膏毒肽原形是尿液中的主要代谢产物,明显高于所有代谢产物的含量。结论本研究将为确定α-鹅膏毒肽在动物体内的残留标志物及应用于α-鹅膏毒肽中毒应急检测与鉴定提供科学依据。
【Abstract】 Objective To detect and identify the metabolites of α-amanitin in rat urine,and to propose the major metabolic pathways of α-amanitin. Methods The rats were administered a single dose of 0. 5 mg/kg α-amanitin by oral gavage. The rat urine samples of administration group and control group were collected respectively. After a simple pretreatment,the urine samples were detected using ultra performance liquid chromatography quadrupole time-of-flight mass spectrometry( UPLC/Q-TOF MS). The data were analyzed by combining with data processing techniques including extracted ion chromatography( EIC) and mass defect filtering( MDF). Results A total of 8 metabolites of α-amanitin,including 5 phase 1 and 3 phase2,were detected in the urine samples. The structures of detected metabolites were elucidated by comparing their characteristic product ions with those of the parent α-amanitin. Based on the identified metabolites,the metabolic pathways of α-amanitin included parent α-amanitin,oxidation,desaturation,decarboxylation and methylation. Among those detected metabolites,the parent α-amanitin was the highest response,much greater than any other α-amanitin metabolites,and it was proposed as the primary metabolites in rat urine. Conclusion The results of the study can provide scientific basis for determining appropriate marker residues of α-amanitin and facilitate to effectively control α-amanitin residues in animals.
