叔丁基对苯二酚对急性矽尘接触后大鼠肺组织血红素加氧酶-1表达的影响

【摘要】 目的研究叔丁基对苯二酚(t BHQ)对急性矽尘接触大鼠肺组织血红素加氧酶-1(HO-1)表达的影响。方法 96只SPF级雄性Wistar大鼠随机分为3组,每组32只。模型组与干预组采用一次性非暴露气管染尘法建立大鼠急性矽尘接触模型,干预组和对照组分别以质量分数1%的t BHQ溶液和1%生理盐水进行干预,1次/d。各组分别于3、14、28、60 d处死8只大鼠。免疫组织化学、蛋白质印迹及实时定量荧光测定各组大鼠肺组织HO-1蛋白的分布、表达及m RNA表达水平。结果与对照组相比,各时间点模型组和干预组HO-1蛋白表达均增强,随着时间延长呈增加趋势,差异有统计学意义(P<0.05),各时间点干预组HO-1蛋白表达高于模型组,除14 d外差异均有统计学意义(P<0.05);与对照组相比,各时间点模型组和干预组HO-1m RNA表达水平增强,随着时间延长呈增加趋势,差异均有统计学意义(P<0.05),各时间点干预组HO-1 m RNA表达水平高于模型组,除3 d外差异均有统计学意义(P<0.05)。结论 HO-1蛋白在急性矽尘接触后大鼠肺组织中的表达上升,t BHQ对其表达具有一定的提升作用。

【Abstract】 Objective To study the expression of HO-1 in lung tissue after rats exposed to acute silica dust. Methods 96 male Wistar rats without specific pathogen(SPF) were randomly separated into three groups,control group, model group and intervention group, 32 rats for each group. Animals in model group and intervention group were exposed to one single unexposed tracheal dust. The intervention group was intervened with 1% t BHQ solution and the control group was given 1%normal saline once a day. At different time points(days 3, 14, 28, and 60), rats in each group were sacrificed for subsequent biochemical analyses(n=8 rats per group and time point).The distribution of HO-1 in lung tissue of each group was detected by immunohistochemistry. The expressions of HO-1 in lung tissues of rats were detected by Western blot method. The expression levels of HO-1 mRNA in lung tissue of rats were detected by RT-PCR. Results Compared with control group,Ho-1 protein expression was enhanced in both the time point model group and the intervention group,and it showed an increasing trend with time, the difference was statistically significant(P<0.05). The expression of the HO-1 in the lungs of rats was higher than that of the model group, except for the 14 th day, the difference was statistically significant(P <0.05).Compared with control group, Ho-1 Mrna expression was enhanced in both the time point model group and the intervention group,and it showed an increasing trend with time, the difference was statistically significant(P<0.05). The expression of the HO-1 mRNA in the lung tissue of the intervention group was higher than that of the model group at each time point. There was a statistically significant difference except for the 3 rd day(P<0.05). Conclusion The expression of HO-1 in lung tissue increase after acute silica exposure in rats.
