硅酸盐细菌代谢产物对矽肺防治作用的初步探索
Effect of silicate bacterial metabolites on prevention and treatment of silicosis
【摘要】 目的探讨硅酸盐细菌代谢产物防治矽肺的可行性。方法 80只SD大鼠,随机分为正常对照组5只、模型对照组35只(分为7个亚组,每亚组5只)、治疗组35只(分为7个亚组,每亚组5只);空白治疗组5只。正常对照组在正常环境中喂养;模型对照组于HOPE-MED 8050动式染毒柜中进行二氧化硅染尘,同时喂养普通饲料;治疗组的二氧化硅染毒方法等同模型对照组,但染毒后将硅酸盐细菌代谢产物原液经雾化后吸入给药,各亚组每次雾化给药30 ml。在实验第1d、3d、1w、2w、4w、8w、12w处死模型对照组和治疗组各亚组,在实验开始后第12w处死正常对照组和治疗对照组,对所有动物进行大体解剖和肺部组织学观察。结果模型对照组和治疗组体重增长均较正常对照组缓慢,但治疗组体重增长优于模型对照组。组织学观察可见模型对照组大鼠矽肺严重,光镜检查见肺内大量泡沫细胞;治疗组病变较轻,光镜下检查见肺内泡沫细胞,但相比模型对照组较少。结论硅酸盐细菌代谢产物可以减轻二氧化硅造成的肺部损伤。
【Abstract】 Objective To explore the feasibility of using silicate bacterial metabolites to prevent and treat experimental silicosis. Methods 80 SPF grade male rats with weights from 140 to 160 g were randomly divided into normal control group( n= 5),model control group( n = 35),treatment group( n = 35) and blank treatment group( n = 5). The model control group and treatment group were divided into 7 subgroups i. e. 1 d,3 d,1 w,2 w,4 w,8 w,and 12 w group,5 rats in each group. The normal control group was fed in normal environment; model control group was exposed to silica dust in HOPE-MED 8 050 dynamic exposure cabinet and fed with normal feed; treatment group was treated with the same method as the model control group,while with silicate bacterial metabolite solution by atomized administration,each subgroup administration of 30 ml each atomization after exposure. Model control group and treatment group were sacrificed at 1 d,3 d,1 w,2 w,4 w,8 w and 12 w respectively. Normal control group and blank treatment group were also sacrificed 12 weeks after the start of the experiment. Gross anatomy and histology of lung were observed in all animals. Results Compared with the normal control group,the body mass growth of the model control group and treatment group was slower,but the weight gain of the treatment group was better than that of the model control group. Histological observation showed that model control group rats were severe silicosis,and light microscopy found a large number of foam cells in the lungs.,The lesions were lighter in the treatment group,and the foam cells in the lung were detected under light microscope,but less than those in the model control group. Conclusion The silicate bacteria metabolites can alleviate the lung injury caused by silica.
