大分子拥挤试剂对细胞外基质作用机制及尚待证实的不利因素
Macromolecular crowding agents for the extracellular matrix: mechanisms and unfavorable factors to be comfirmed
【摘要】 背景:大分子拥挤试剂对细胞外基质成分中的胶原蛋白、糖胺聚糖、生长因子等都会产生一定的影响。目的:总结大分子拥挤试剂对细胞外基质的影响,以及这些大分子拥挤试剂在各种细胞或组织中的应用。方法:应用计算机检索Pub Med、NCBI、中国知网数据库2001至2017年的文献,中、英文检索关键词为"Macromolecular crowding,MMC,Tissue Engineering,Collagen;大分子拥挤,细胞外基质",总结大分子拥挤试剂在细胞外基质中及组织工程中的应用进展。结果与结论:大分子拥挤试剂可通过排斥容积效应增加细胞外基质成分的沉积,如胶原、糖胺聚糖及生长因子等,促进成骨细胞的黏附、迁移、生长和分化,促进骨组织工程中再生医学的发展。但是,大分子拥挤试剂还有很多尚未被发现或证实的作用及作用机制,将其应用到人体是否会产生毒性作用还尚未证实,这些问题都需要大量的实验研究去证实。
【Abstract】 BACKGROUND: Macromolecule crowding agents exert a certain effect on collagen, glycosaminoglycans and growth factors in the extracellular matrix. OBJECTIVE: To summarize the effects of macromolecule crowding agents on the extracellular matrix and their applications in various cells or tissues. METHODS: "Macromolecular crowding, MMC, Tissue Engineering, Collagen" were used as key words to retrieve articles addressing the application of macromolecular crowding agents in the extracellular matrix and in the tissue engineering in Pub Med, NCBI, CNKI from 2001 to 2017. RESULTS AND CONCLUSION: Macromolecule crowding agents can increase the deposition of extracellular matrix components, such as collagen, glycosaminoglycans and growth factors, to promote osteoblast adhesion, migration, growth and differentiation, as well as to promote the development of regenerative medicine in bone tissue engineering. However, some roles and mechanisms of macromolecule crowding agents have yet to be found or confirmed, and whether they can produce toxic effects in the human body has yet to be confirmed. Therefore, further experimental research on these problems is warranted.
