电针刺激脓毒血症大鼠足三里穴效果评价
The Impact Evaluation of Rats with Sepsis by Electrical Stimulation Therapy on Zusanli Point
【摘要】 目的结合动物行为学及免疫组化方法探讨电针足三里穴对脓毒血症盲肠结扎穿刺术(cecal ligation and puncture,CLP)模型大鼠的效果。方法根据经典文献采用盲肠结扎穿孔法制作CLP模型大鼠,随机分为模型组、假手术组和治疗组,每组各20只。治疗组每天给予足三里穴电刺激治疗,每天2次,持续1周,模型组和假手术组不做干预治疗。同时记录各组大鼠1周内死亡率和生存时间;于试验结束时取外周血,采用ELISA双抗体夹心法检测TNF-α、MCP-1的活性表达水平。结果电针治疗组大鼠生存时间显著提高,死亡率显著下降,死亡率低于模型组,生存时间高于模型组,差异有统计学意义(P<0.05,P<0.01);电针治疗组大鼠外周血中TNF-α、MCP-1的活性表达水平显著低于模型组(P<0.05,P<0.01)。结论足三里穴电针法可以对延长CLP大鼠的生存时间,降低死亡率具有较好的作用,并可降低外周血中TNF-α、MCP-1的含量。
【Abstract】 Objective Through animal behavioral science and immunohistochemical method, this paper tried to explore the effects of electro-acupuncture on CLP rats. Methods According to the literature, with cecal ligation and puncture operation, CLP modle was established, the rats were randomized into model group, sham operation group and treatment group, 20 in each group. The treatment group was daily given high-frequency electrical stimulation therapy on Zusanli point, twice daily for one week, model group and sham operation group without intervention. At the same time recorded the mortality and survival time within one week; Peripheral blood was taken at the end of the test, ELISA double antibody sandwich assay was used to detect the expression level of TNF-α and MCP-1. Results After giving electrical stimulation therapy on Zusanli point of CLP rats, the survival time of rats in treatment group increased and the mortality rate significantly decreased, the mortality rate was lower than that of the model group, and the survival time was higher than that of the model group, the mortality rate was lower than that of the model group, the differences are statistically significant(P<0.05,P<0.01). TNF-α and MCP-1 like immunoreactive intensity in peripheral blood of treatment group were stronger than that of the model group and the there was significant difference(P<0.05,P<0.01).Conclusion The electrical stimulation therapy on Zusanli point has good effect on prolonging the survival time and reducing mortality of CLP rats, and could decrease the content of TNF-α and MCP-1 in peripheral blood.
