三种浓度瘦素对大鼠自体颗粒脂肪移植的研究
The Study of Fatty Transplantation in Rats with Three Levels of Leptin
【摘要】 目的实验研究三种浓度瘦素对大鼠颗粒脂肪移植30天后成活情况的效果。方法用生理盐水及l0ng/mL、30ng/mL、50ng/mL三种浓度瘦素处理大鼠自体颗粒脂肪,在30天后取出移植组织称重,行HE染色,观察移植组织成活情况。实验数据使用SPSS18.0统计软件进行处理。结果移植组织重量维持率在移植30天后,NS处理组为45%;l0ng/mL瘦素组为67.5%;30ng/mL瘦素组为75%;50ng/mL瘦素组为77.5%。除30ng/mL瘦素组与50ng/mL瘦素组间差异无显著性意义外,其余各组间重量维持率相比,差异有显著性意义(P<0.05)。用瘦素处理的脂肪组织成活较好。结论在0~50ng/mL范围内,增加瘦素的浓度能提高大鼠自体颗粒脂肪移植组织的移植成活率。但瘦素浓度大于30ng/mL后对移植脂肪成活影响可能减弱。
【Abstract】 Objective To study the effect of three levels of leptin on the growth of fat removal of rat particles in 30 days. Methods With physiological saline and l0 ng/mL/30 ng/mL and 50 ng/mL, three concentrations of leptin deal with autologous fat granules in rats, weight the transplant tissue after 30 days, HE staining, and observe the graft survival. The experimental data was processed using SPSS18.0 statistical software. Results To detection the migration tissue weight maintenance rate after 30 days, The sodium chloride treatment group was 45% The l0 ng/mL leptin group was 67.5%; The 30 ng/mL leptin group was 75%; The 50 ng/mL leptin group was 77.5%. In addition to the difference between the 30 ng/mL leptin group and the 50 ng/mL leptin group, the difference in weight maintenance was significant(P<0.05). Fat tissue treated with leptin is better. Conclusion In the range of 0 ~ 50 ng/mL, increasing the concentration of leptin increases the survival rate of the transplanted tissue of the rat. But the concentration of leptin is greater than 30 ng/m L and the effect on the transplanted fat may be attenuated.
