食管鳞癌中miR-204、miR-205相关ceRNA网络构建及部分lncRNA功能分析

【摘要】 目的用生物信息学方法预测食管鳞癌(esophageal squamous cell carcinoma,ESCC)组织中miR-204、miR-205相关竞争性内源RNA(ceRNA)网络。方法通过TCGA数据库筛选癌及癌旁组织差异表达的mRNA、长链非编码RNA(lncRNA)和miRNA,构建miR-204、miR-205相关ceRNA网络,并对筛选出的差异基因进行生存分析,利用GO、KEGG和PPI进一步分相关lncRNA的功能。结果以log FC>2且P<0.05为标准,筛选出和miR-204、miR-205相关差异表达mRNA 13个,lncRNA 38个,并构建了lncRNA-miRNA-mRNA调控网络。LINC00504、FER1L6-AS1与亚洲人ESCC患者生存相关。GO、KEGG和PPI分析显示:7个同时调节miR-204、miR-205的lncRNA可能参与ESCC的形成和进展。结论通过分析ESCC转录组测序数据,预测出以miR-204和miR-205为节点的调控网络,可能是ESCC重要的调控机制和诊疗靶点。

【Abstract】 Objective To predict miR-204,miR-205 related ceRNA regulatory network of esophageal squamous cell carcinoma( ESCC) by on-line database. Methods Screening of differentially expressed mRNAs,lncRNAs and miRNAs in ESCC and adjacent non-tumor esophageal tissues were performed according to transcriptome data of TCGA.The miR-204 and miR-205 related ceRNA regulatory network was constructed. Survival,GO,KEGG and protein-protein interwork( PPI) analysis of the lncRNAs and mRNAs in the ceRNA netwrk were performed. Results Thirty eight differentially expressed lncRNAs,13 differentially expressed mRNAs relate to miR-204,miR-205 in ESCC were screened out from TCGA database. ceRNA network was constructed. There were 7 lncRNAs which interacted with miR-204 and miR-205 simultaneously,and might be involved in the formation and progression of ESCC by GO,KEGG and PPI analysis. LINC00504 and FER1 L6-AS1 were related to overall survival of ESCC of Asian patients. Conclusion By analyzing the transcriptome data of ESCC in on-line database,we predict the ceRNA regulatory network of miR-204 and miR-205,which may be an important regulatory mechanism and target for diagnosis and treatment of ESCC.
