香芩解热颗粒对2,4-二硝基苯酚致热大鼠的解热作用研究

【摘要】 目的:观察香芩解热颗粒对非感染性动物模型的解热作用并探讨其机制。方法:将进行体温筛选后的大鼠随机分为正常组、模型组、布洛芬组(阳性对照组)以及香芩解热颗粒组(XQ组)。除正常对照组外,其余各组背部皮下注射2,4-二硝基苯酚,复制大鼠非感染性发热模型,造模前预防性给药两次(间隔0.5 h),监测造模后0.5、1、2、3 h肛温,应用酶联免疫法(ELISA)测定大鼠血清IL-1β(白细胞介素-1β)、TNF-α(肿瘤坏死因子α)及下丘脑PGE2(前列腺素E2)、c AMP(环磷酸腺苷)含量,应用PCR技术测定下丘脑PGE2受体EP1、EP4 mRNA相对表达量。结果:模型组大鼠在造模后的0.53 h内体温均显著高于正常对照组(P<0.05),造模0.5 h达到峰值;与模型组比较,造模0.5 h后香芩解热颗粒组大鼠体温均显著降低(P<0.05)。ELISA结果显示:与正常对照组相比,模型组大鼠血清IL-1β、TNF-α及下丘脑PGE2、c AMP含量显著上升;与模型组比较,布洛芬组大鼠血清IL-1β、TNF-α及下丘脑PGE2、c AMP值显著下降,但香芩解热颗粒组仅下丘脑PGE2、c AMP值显著下降。PCR结果显示:与正常对照组比较,模型组EP1mRNA、EP4mRNA相对表达量显著上升,与模型组比较,香芩解热颗粒组EP1mRNA、EP4mRNA显著下降,布洛芬组EP1mRNA显著下降。结论:香芩解热颗粒对酵母型发热大鼠有明显的退热趋势,其退热作用可能与降低下丘脑体温正调节介质PGE2、c AMP的含量及下调下丘脑EP1mRNA、EP4mRNA表达有关。

【Abstract】 Objective:To investigate the antipyretic effect of Xiangqin Jiere Granule on 2,4-dinitrophenol induced febris rats.Methods:40 rats were selected and radomly divided into normal group,model group,Xiangqin Jiere Granule group and ibuprofen group.Prophylactically and respectively intragastric administrated each group with distilled water,17 g/kg Xiangqin Jiere Granule and 2.52 m L/kg ibuprofen.0.5 hours later,intragastric administrated again and established fever model by sc giving 2 mg/kg 2,4-dinitrophenol on the back,except for the normal group.The anal temperature at 0.5,1,2,3 h,the serum level of IL-1β and TNF-α,and the level of PEG2,c AMP,EP1 mRNA,EP4 mRNA in hypothalamus were all detected.Results:The body temperater of model group rats were gradually increased,and reached peak at 0.5 h.Compare with the model group,Xiangqin Jiere Granule can significantly decreased the temperature of febris rats( P < 0.05).The ELISA and PCR results indicated that Xiangqin Jiere Granule can significantly decreased the level of PEG2 and c AMP in hypothalamus,as well as the EP1 mRNA and EP4 mRNA expressions in hypothalamus( P < 0.05),but not the serum level of IL-1β and TNF-α.Conclusion:Xiangqin Jiere Granule has a good antipyretic activity to p-nitrophenol-induced febris rats.Its mechanism may be related to the inhibition of hypothalamic signal molecule( PEG2 and c AMP) and the down-regulation of EP1 mRNA and EP4 mRNA expressions in hypothalamus.
