目的:研究壳聚糖(CS)及壳聚糖负载的一氧化氮(NO)、胰岛素样生长因子-1C(IGF-1C)与人胎盘来源的间充质干细胞(hP-MSCs)共培养后对细胞功能的影响。方法:利用可稳定表达绿色荧光蛋白(GFP)和萤火虫荧光蛋白(Fluc)的hP-MSCs与3种不同的生物活性材料水凝胶共培养,通过MTT、生物发光成像技术以检测水凝胶对细胞增殖的影响,同时利用real time PCR检测水凝胶对细胞增殖、凋亡、抗炎及血管新生相关细胞因子的表达作用。结果:CS-IGF-1C水凝胶可明显促进hP-MSCs的增殖及上调相关基因IGF-1(11.51)、HGF(6.64)、EGF(9.78)的表达(P<0.05),抑制hP-MSCs的凋亡并下调相关基因BAX/BAD(1.22/1.12)、FAS/FASLG(1.31/1.70)的表达(P<0.05),同时还可显著上调hP-MSCs对TSG-6(11.61)及PGE2(16.04)的表达(P<0.05)。而CS-NO可显著促进hP-MSCs对VEGF(13.28)、PDGF(8.49)的表达作用(P<0.05)。结论:不同水凝胶对MSCs的影响不同,CS-IGF-1C水凝胶...
