siRNA干扰技术在耐药性卵巢癌中的研究进展

【摘要】 卵巢癌死亡率居女性生殖系统恶性肿瘤之首,卵巢癌起病隐匿,早期无特殊临床表现,多数患者在晚期时才被发现,经肿瘤细胞减灭术及术后辅以铂类为基础的化疗后,部分患者出现病情缓解,但仍有一部分患者表现出化疗耐药以至于病情进展或复发。RNA干扰是由双链RNA介导的序列特异性的转录后基因沉默现象,小干扰RNA在RNA干扰过程中发挥核心作用。文章就小干扰RNA技术原理、递送及其近5年在耐药性卵巢癌方面的研究进展进行综述。

【Abstract】 Ovarian cancer has the highest mortality rate among female reproductive cancer. Because it has no special clinical manifestation in early stage,and the majority of patients are diagnosed in advanced stage. This is a disease with a propensity to recur or deteriorate despite dramatic responses to the treatment of a combination of cytoreductive surgery and platinum-based chemotherapy. RNA interference is a sequence-specific posttranscriptional gene silencing triggered by double-stranded RNA,in which small interfering RNA plays a core role. This paper reviewed the research progress on small interfering RNA technical principle,delivery,and its application in drug-resistant ovarian cancer in recent five years.
