刺五加提取物联合运动改善健康SD大鼠骨骼肌功能性抗交感的实验研究
Experimental Study on Extract of Acanthopanax Senticosus Combined with Exercise Improved Functional Sympatholysis of Skeletal Muscle in Healthy SD Rats
【摘要】 目的:观察8周刺五加提取物联合运动对骨骼肌功能性抗交感的影响并探讨一氧化氮合酶(NOS)/一氧化氮(NO)系统在其间的可能作用机制。方法:60只健康雄性SD大鼠随机分为安静对照组(S)、刺五加组(A)、运动组(E)以及运动+刺五加组(EA),干预时间为8周。实验结束后麻醉动物,电刺激腰部交感神经干和坐骨神经分别诱导缩血管反应以及小腿三头肌收缩,记录静息状态下和肌肉收缩过程中交感神经电刺激(2和5 Hz)时的股动脉电导(FVC),功能性抗交感用肌肉收缩时FVC对交感神经电刺激反应的变化率(%FVC,即交感缩血管反应)与静息状态下的差值表示,即△%FVC=%FVC肌肉收缩-%FVC静息。分离比目鱼肌(Sol)和腓肠肌(Gas),测定一氧化氮(NO)含量以及神经元型NOS(n NOS)、内皮型NOS(e NOS)和诱导型NOS(i NOS)活性。结果:(1)%FVC:静息状态下,A组%FVC低于S组(P<0.05);肌肉收缩时,E组%FVC低于S组(P<0.05);肌肉不同状态下EA组%FVC均显著性下降(P<0.05)。(2)Δ%FVC:2 Hz交感电刺激时,E组△%FVC高于S组(P<0.05),EA组高于A组和S组(P<0.05);5 Hz交感电刺激时,A组和E组△%FVC均高于S组(P<0.05),EA组高于其他3组(P<0.05)。(3)NO含量:A组和E组高于S组(P<0.05),EA组高于其他3组(P<0.05)。(4)NOS活性:i NOS在不同肌纤维类型间以及各组间均无显著性差异(P<0.05);e NOS不存在肌纤维类型依赖性,Gas和Sol中e NOS均为A组高于S组(P<0.05),EA组高于E组和S组(P<0.05);n NOS在各组Gas均高于Sol(P<0.05),E组Gas中n NOS高于S组(P<0.05),EA组高于A组和S组(P<0.05)。结论:刺五加提取物联合运动对改善骨骼肌静息、收缩时的交感缩血管反应和功能性抗交感具有协同效应,其机制可能与e NOS(快肌和慢肌)和n NOS(快肌)活性增加进而上调NO含量有关
【Abstract】 Objective:To observe the effects of extract of Acanthopanax Senticosus combined with aerobic exercise on functional sympatholysis of skeletal mus-cle and investigate the possible mechanism of nitric oxide synthase(NOS)/nitric oxide(NO)system. Methods:Healthy male SD rats(n=60)were randomized tosedentary control(S),Acanthopanax Senticosus(A),exercise-trained(E)or exercise-trained plus Acanthopanax Senticosus(EA)groups and intervention lastedfor 8 weeks. Following completion of the experiments,rats were anaesthetized,and vasoconstriction and triceps surae muscle contraction were induced by elec-trostimulation of lumbar sympathetic chain and sciatic nerve respectively. Femoral vascular conductance(FVC)during sympathetic nerve electrostimulation(2 Hz and 5 Hz)was recorded at rest and during muscle contraction respectively. Functional sympatholysis was represented of difference between change rate ofFVC to sympathetic nerve electrostimulation(%FVC,ie.sympathetic vasoconstriction)during muscle contraction and at rest(△%FVC=%FVCmuscle contrac-tion-%FVCat rest). Soleus muscle(Sol)and gastrocnemius muscle(Gas)were isolated and NO content,and neuronal NOS(n NOS),endothelial NOS(e NOS)and inducible NOS(i NOS)activity were determined. Results:(1)%FVC:at rest,%FVC in A group was lower than S group(P<0.05);during muscle contrac-tion,%FVC in E group was lower than S group(P<0.05);%FVC in EA group was reduced(P<0.05)at any muscle state.(2)△%FVC:during sympathetic nerveelectrostimulation at 2 Hz,△%FVC in E group was higher than S group(P<0.05)and those of EA group higher than A and S groups(P<0.05);at 5 Hz,△%FVC in A and E groups were higher than S group(P<0.05)and those of EA group higher than other three groups(P<0.05).(3)NO content:NO in A and E groups werehigher than S group(P<0.05)while EA group higher than other three groups(P<0.05).(4)NOS activity:there was no significant change in i NOS between musclefiber types and groups(P>0.05);e NOS was independent of muscle fiber types while activity was higher in A than S group(P<0.05)and EA group higher than Eand S groups(P<0.05)in both Gas and Sol;n NOS were higher in Gas than Sol of all groups(P<0.05),n NOS was higher in E than S group of Gas(P<0.05)whileEA group higher than A and S groups(P<0.05). Conclusions:There was synergistic effect of extract of Acanthopanax Senticosus combined with exercise in im-provement of skeletal muscle sympathetic vasoconstriction at rest and during contraction as well as functional sympatholysis,which was related with simultane-ous upregulation of e NOS(fast and slow muscle)and n NOS(fast muscle)and augment of NO.
