针刺对肥胖鼠胸腺、脾脏及肠派氏淋巴结中T淋巴细胞亚群的影响

【摘要】 目的探究针刺对肥胖鼠肠胸腺、脾脏及肠中段派氏淋巴结中的T淋巴细胞亚群的影响。方法将100只C57BL/6小鼠,随机筛选10只作为正常组,其余饲以高脂饮食,8周后,将体重大于正常组均值20%的肥胖鼠均分成模型组和针刺组,观察比较针刺干预前后各组小鼠的体重、Lee’s指数、附睾脂肪质量及重要免疫器官胸腺、脾脏、肠中段派氏淋巴结中的T淋巴细胞亚群CD4+、CD8+、CD4/CD8及CD11c+的含量变化。结果模型组体重、Lee’s指数及附睾脂肪质量均显著高于正常组;模型组脾脏中CD4+、CD4/CD8比值显著低于正常组,肠派氏淋巴结中的CD4+、CD4/CD8比值显著低于正常组,CD8+及CD11c+的含量显著高于正常组。针刺干预后,针刺组体重、Lee’s指数及附睾脂肪质量显著下降,肠派氏淋巴结中的CD4+、CD4/CD8比值显著升高,CD8+及CD11c+的含量显著降低。结论肥胖鼠存在一定程度的免疫功能紊乱,主要表现在肠粘膜免疫紊乱或低下,通过针刺治疗调整后可以有效改善,揭示针灸减肥调整肠黏膜免疫的机制。

【Abstract】 Objective: To investigate the effect of acupuncture on T lymphocyte subsets in thymus,spleen and peyer’ patches of obese mice. Methods 10 of 100 C57 BL/6 mice were randomly selected as normal group and the other were fed with high fat diet. After 8 weeks,the obese mice with body weight were greater than 20% average value of the normal group. Then,these were divided into model and acupuncture group. Compared the difference of weight,Lee’s index,epididymal fat mass and T lymphocyte subsets( CD4+,CD8+,CD4/CD8 and CD11c+) of thymus,spleen and intestinal peyer’patches. Results The body weight,Lee’s index and epididymal fat mass in the model group were significantly higher than in the normal group. The content of CD4+ and ratio of CD4/CD8 of spleen in the model group was significantly lower than that in the normal group; the content of CD4+ and the ratio of CD4/CD8 of peyer’ patches in the model group was significantly lower than that in the normal group,and the content of CD8+ and CD11c+ in the model group were significantly higher than those in the normal group. After acupuncture treatment,the weight,Lee’s index and epididymal fat mass were significantly decreased,and the content of CD4+and the ratio of CD4/CD8 of peyer’ patches were significantly increased,and the contents of CD8+and CD11 c+were significantly decreased. Conclusion There is a certain degree of immune dysfunction in obese mice,mainly manifested in the immune disorder of the intestinal mucosa.Through the acupuncture treatment can be adjusted to improve the effectiveness and reveal the mechanism of acupuncture and moxibustion to adjust intestinal mucosal immunity.
