蜂毒素体外抑制小鼠结肠癌CT-26细胞的实验研究
Research on effect of Melittin in vitro inhibition of mouse colon cancer CT-26 cells
【摘要】 目的探讨蜂毒素对小鼠结肠癌CT-26细胞的迁移和凋亡的影响及其分子机制。方法 MTT法检测蜂毒素对小鼠结肠癌CT-26细胞增殖的影响;Transwell小室实验检测蜂毒素对CT-26细胞迁移能力的影响;流式细胞技术检测蜂毒素对CT-26细胞凋亡的影响;Western Blot检测经蜂毒素作用后的CT-26细胞中Caspase-9,Caspase-3和PARP的蛋白水平。结果 Transwell小室实验显示,经1μg/ml、2μg/ml蜂毒素处理过的CT-26细胞穿膜数明显减少并呈一定的浓度依懒性(P<0.01)。流式细胞技术检测显示,经1μg/ml、2μg/ml蜂毒素处理过的CT-26细胞凋亡率明显提高并呈一定的浓度依懒性(P<0.01)。Western Blot结果显示,经1μg/ml、2μg/ml蜂毒素处理过的CT-26细胞,Caspase-9,Caspase-3和PARP的蛋白的表达水平明显降低(均P<0.01)。结论蜂毒素能明显抑制小鼠结肠癌CT-26细胞的增殖与迁移,促使细胞的凋亡,其机制可能与下调Caspase-9,Caspase-3和PARP蛋白水平直接相关。
【Abstract】 Objective To investigate the effect of melittin on the migration and apoptosis of mouse colon cancer CT-26 cells and its molecular mechanism. Methods The effect of melittin on the proliferation of colon cancer CT-26 cells was detected by MTT assay. The effect of melittin on the migration of CT-26 cells was detected by Transwell chamber test. The effect of melittin on CT-26 cells was detected by flow cytometry The expression of Caspase-9,Caspase-3 and PARP in CT-26 cells treated with melittin was detected by Western Blot. Results Compared with the negative control group,mice colon cancer treated with different concentrations of melittin CT-26 cells in a dose-dependent growth inhibition( P < 0. 01). Transwell chamber experiments showed that the number of CT-26 cells treated with 1μg/m L,2μg/m L melittin was significantly reduced and showed a certain concentration( P < 0. 01). Flow cytometry showed that the apoptosis rate of CT-26 cells treated with 1μg/m L and 2μg/m L melittin was significantly increased and decreased at a certain concentration( P < 0. 01). Western Blot showed that the expression levels of Caspase-9,Caspase-3 and PARP in CT-26 cells treated with 1μg/m L,2μg/m L melittin were significantly decreased( all P < 0. 01). Conclusion Melittin can significantly inhibit the proliferation and migration of mouse colon cancer CT-26 cells and promote the apoptosis of cells. The mechanism may be related to down-regulation of Caspase-9,Caspase-3 and PARP protein levels.
