用多糖衍生物类手性固定相分离5种β2-受体激动剂药物对映体
Enantioseparation of five β2-receptor agonists via polysaccharide derivatives chiral stationary phase
【摘要】 目的建立分离克仑特罗、妥洛特罗、特布他林、氯丙那林及丙卡特罗5种药物对映体的高效液相色谱法。方法采用HPLC法,用Chiralpak ID手性色谱柱进行分离,考察了酸碱添加剂的种类、浓度;有机改性剂的种类、浓度等因素对分离度的影响,优化了分离条件。结果通过分离条件的优化,确定了分离5种β2-受体激动剂药物对映体的最佳条件。在最佳色谱条件下,克仑特罗、妥洛特罗、氯丙那林和丙卡特罗得到了完全分离,特布他林对映体达到了部分分离。结论 Chiralpak ID手性色谱柱适于分离除特布他林以外的4种β2-受体激动剂药物对映体。
【Abstract】 Objective To establish a high performance liquid chromatography method for the chiral separation of clenbuterol,tulobuterol,terbutaline,clorprenaline and procaterol. Methods With the application of Chiralpak ID column,the factors influencing the chiral separation including the type and concentration of organic modifier,the addition of acid or alkaline additive were investigated. And the chromatographic separation conditions were optimized. Results The optimum conditions for the enantioseparation of five β2-receptor agonists were determined. Under the optimal mobile phase,clenbuterol,tulobuterol,clorprenaline and procaterol were completely separated,while terbutaline was partially separated. Conclusion Chiralpak ID column is suitable for the separation of four β2-receptor agonists except terbutaline.
