洋水仙Dutch Master愈伤组织的诱导、分化及植株再生
Callus Induction, Differentiation and Plant Regeneration of Narcissus pseudonarcissus cv. Dutch Master
【摘要】 以洋水仙品种Dutch Master的鳞茎、叶、花葶及无菌苗叶片作为外植体,选用MS为基本培养基,分别添加不同质量浓度的NAA(0.5,1.0,1.5mg·L-1)、IBA(0,0.5,1.0mg·L-1)、2,4-D(0,0.1,0.2,0.5,1.0,2.0mg·L-1)、6-BA(0.5,1.0,1.5,2.0,3.0mg·L-1)和KT(0.1,0.3,0.5mg·L-1),研究不同外植体和不同外源激素配比对小鳞茎的诱导,愈伤组织的诱导、继代及植株再生的影响。结果表明:在Dutch Master初代培养阶段,6-BA的影响效应最大,随着6-BA质量浓度的升高,诱导率呈上升趋势,最适宜的初代培养基配比为:MS+6-BA 3.0mg·L-1+NAA 0.5mg·L-1+2,4-D 0.2mg·L-1;以带鳞茎盘的双鳞片作为外植体诱导小鳞茎效果最好,诱导率达76.7%;鳞片部位以鳞茎外层鳞片的培养效果最好,分化率达97.5%;在Dutch Master愈伤组织诱导阶段,2,4-D的影响效应最大,当2,4-D浓度为1mg·L-1时,愈伤组织诱导率的均值(K2)为峰值,最适宜的愈伤组织诱导培养基配比为:MS+2,4-D 1mg·L-1+6-BA 1.5mg·L-1+KT 0.1mg·L-1,Dutch Master无菌苗叶片愈伤组织诱导率可以达到81.3%;在Dutch Master愈伤组织继代培养阶段,最适宜的继代培养基配比为:MS+2,4-D 0.5mg·L-1+6-BA 1mg·L-1,可以增殖分化得到没有玻璃化的小植株。
【Abstract】 The effects of different explants and exogenous plant growth regulators have been studied on bulblet induction, callus induction, subculture and plant regeneration. Bulbs, leaves, scapes and tube-plant leaves of Narcissus pseudonarcissus cv. Dutch Master were used as explants, and MS media was used as the basic media, supplemented with different mass concentrations of NAA(0.5,1.0,1.5 mg·L-1), IBA(0,0.5,1.0 mg·L-1), 2,4-D(0,0.1,0.2,0.5,1.0,2.0 mg·L-1), 6-BA(0.5,1.0,1.5,2.0,3.0 mg·L-1) and KT(0.1,0.3,0.5 mg·L-1)respectively. The effect of 6-BA was the greatest in the initial culture stage of Dutch Master. With the increase of 6-BA concentration, the induction rates increased. The optimizing medium for initial culture was MS + 6-BA 3.0 mg·L-1+ NAA 0.5 mg·L-1+ 2,4-D 0.2 mg·L-1. The results showed that twin-scale with basal plate was more suitable explant,the induction rate was 76.7%, and the cultivation effect of external layer scale was better, with differentiation rate of 97.5%. The effect of 2,4-D was the greatest in the callus induction stage of Dutch Master. When the concentration of 2,4-D was 1 mg·L-1,the mean value of callus induction rate(K2) was the peak. The callus induction rate of tube-plant leaves reached 81.3% on the media of MS + 2,4-D 1 mg·L-1+ 6-BA 1.5 mg·L-1+ KT 0.1 mg·L-1. The callus grew further and regenerated no vitrified plantlets on the media of MS + 2,4-D 0.5 mg·L-1+6-BA 1 mg·L-1.
