护骨素和κB-受体激活因子水平变化对缺血性股骨头坏死保护作用效果及作用机制分析

【摘要】 目的探讨护骨素(OPG)和κB-受体激活因子(RAN)水平对缺血性股骨头坏死保护作用效果及其作用机制。方法回顾性选取2013年2月至2017年3月北京市昌平区中医医院病理科保存的缺血性股骨头坏死组织33例作为观察组,同时选取11例股骨颈骨折组织作为对照组,采用实时荧光定量PCR和Western blotting法检测两组OPG和RAN mRNA和蛋白表达水平,比较两组不同性别、不同月经状态患者OPG、RAN表达的差异。结果观察组OPG mRNA和RAN mRNA相对表达量分别为4.32±0.41和0.83±0.15,明显高于对照组(P<0.05);观察组和对照组组织中均未检测到RAN蛋白表达;观察组和对照组OPG蛋白相对表达量差异比较,差异无统计学意义(P>0.05);观察组男性和女性患者OPG mRNA和蛋白、RAN mRNA表达比较,差异无统计学意义(P>0.05);观察组未绝经和已绝经女性OPG mRNA和蛋白相对表达量比较,差异无统计学意义(P>0.05);观察组已绝经女性RAN mRNA相对表达量(1.06±0.22)明显高于未绝经女性(0.71±0.16),差异具有统计学意义(P<0.05)。结论 OPG和RAN在缺血性股骨头坏死组织中可能具有一定的作用,其中女性月经状态对RAN表达有一定的影响。

【Abstract】 Objective To investigate protective effect and mechanism of osteoprotegerin( OPG) and receptor activator κB-( RAN) in avascular necrosis organization of the femoral head. Methods From February 2013 to March 2017,33 cases of ischemic necrosis of femoral head were preserved in Beijing Changping Hospital of Traditional Chinese Medicine( as observation group,and 11 cases of femoral neck fracture were selected as control group,OPG and RANK mRNA and protein expression were detected by real time fluorescence quantitative PCR and Western blotting; The differences in the expression of OPG and RAN in two groups of patients with different sex and different menstrual state were compared. Results The relative expression of OPG mRNA and RAN mRNA in the observation group were( 4. 32 ± 0. 41) and( 0. 83 ± 0. 15),which were significantly higher than that in the control group( P < 0. 05). The expression of RAN protein was not detected in the observation group and the control group; there was no significant difference in the relative expression of OPG protein between the observation group and the control group( P > 0. 05). There was no significant difference in the expression of OPG mRNA,protein and RAN mRNA in the men and women of the observation group( P > 0. 05). There was no significant difference in the relative expression of OPG mRNA and protein between the non menopause and the menopause women in the observation group( P > 0. 05). In the observation group,the relative expression of RAN mRNA in menopausal women was( 1. 06 ± 0. 22),which was significantly higher than that of non menopausal women( 0. 71 ± 0. 16)( P < 0. 05). Conclusion OPG and RAN may play a role in ischemic necrosis of the femoral head,and the menstrual state of women has a certain effect on the expression of RAN.
