粘柄丝膜菌多糖的提取工艺及抗氧化活性
Extraction and Antioxidant Activities of Polysaccharides from Cortinarius collinitus Fruiting Bodies
【摘要】 采用响应面法优化粘柄丝膜菌(Cortinarius collinitus)子实体多糖的超声辅助提取工艺,分别采用高效液相色谱(high performance liquid chromatography,HPLC)和气相色谱(gas chromatography,GC)进行多糖样品分子量测定和单糖组成分析;并检测多糖样品体外清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基和羟自由基的活性。结果表明:最佳提取工艺条件为液料比40∶1,提取温度68℃,提取时间2.6h,超声功率300 W,在此条件下粘柄丝膜菌多糖得率为5.97%,较常规水提(4.73%)提高了26%;粘柄丝膜菌多糖中糖含量为72.35%,该多糖是由鼠李糖、阿拉伯糖、木糖、甘露糖、半乳糖和葡萄糖6种单糖构成的杂多糖;800μg/mL多糖对DPPH自由基的清除率达71%,4mg/mL多糖对羟自由基的清除率达63%。
【Abstract】 Polysaccharide extraction from fruiting bodies of Cortinarius collinitus was optimized by the response surface methodology,and then the extract was determined for relative molecular weight and monosaccharide compositions by high performance liquid chromatography(HPLC)and gas chromatography(GC),respectively.Finally the extracted polysaccharide was analyzed for free radical scavenging activities.The optimized conditions for ultrasonic wave-assisted extraction were as follows∶liquid/solid ratio∶40∶1(mL∶g),extraction temperature:68 ℃;extraction time:2.6 h;and ultrasonic wave power:300 W.Extractions done under these conditions resulted in an average polysaccharide yield of 5.97%,which was a 26%increase in polysaccharide yield compared with that of the traditional water extraction method(4.73%).The extracted polysaccharide consisted of rhamnose,arabinose,xylose,mannose,galactose,glucose.Seventy percent of the free radical 1,1-diphenyl-2-picrylhydrazyl(DPPH) was scavenged by 800 μg/mL polysaccharides,and 63% of the·OH free radical was scavenged by 4 mg/mL polysaccharides.
