MRI动态监测前列腺癌动物模型中瘤组织生长的实验研究

【摘要】 目的 探讨3.0T MRI动态监测小鼠前列腺癌模型中瘤组织生长的可行性及其应用价值。方法 采用瘤组织块原位种植法建立C57BL/6小鼠前列腺原位种植瘤模型15只。接种后第3、6、9、12天用MR对模型鼠扫描。第12天最后1次扫描结束,将存活模型鼠处死并取出瘤组织,游标卡尺测量瘤组织最大长径,测量结果和MRI测量值进行比较,组织行病理学检查。结果除去因手术损伤、感染、MR扫描过程中意外死亡4只小鼠,其余11只鼠均建模成功。MRI第6天开始成功监测到模型鼠前列腺肿瘤的部位与大小,可检测到肿瘤的最小长径为2.70 mm,敏感性>90%。MRI和游标卡尺测量肿瘤最大长径比较无显著差异(P>0.05),两者相关性为0.996(P<0.01)。病理结果可见典型的肿瘤细胞,淋巴管内可见癌栓。结论 MRI可早期明确小鼠前列腺原位种植瘤模型肿瘤组织的形态、部位及大小,可为后续前列腺动物模型的化疗药物研究提供良好的影像学监测手段。

【Abstract】 Objective To explore the feasibility and application of the 3.0 T MRI by dynamicly monitoring tumor tissue growth in mice prostate cancer model.Methods C57BL/6 prostate carcinoma mice(n=15) were modeled by the tumor tissue in-situ implantation.Those model mice were scanned using the 3.0T MR every 3 days after explantation until the 12th day.All model mice were killed,the tumor tissue were taken out and the largest diameters of tumors were measured and compared with the results measured using MRI,pathological examination was performed.Results 11 mice were successfully modeled and 4 mice were died of surgical injury,infection or accidence during MR scanning.On the 6th day,the position and size of prostate carcinoma could be detected successfully by MRI,and the minimum length of diameter was 2.70 mm with sensitivity>90%.There was no significant difference between MRI and calipers in measuring the maximum diameter of tumors outside the body(P>0.05),and the Pearson correlation was 0.996(P <0.01).The pathological results showed typical tumor cells,and the tumor suppositories were visible in the lymphatic vessels.Conclusion MRI can clearly define the morphology,position and size of in-situ prostatic implant tumor in mice,which could provide a favorable imaging monitoring method for subsequent prostate cancer animal models with chemotherapy.
