【摘要】 目的探讨girdin蛋白表达与胃癌的临床病理特征及患者预后的关系和临床意义。方法采用免疫组织化学和蛋白质印迹法检测girdin蛋白在人胃癌细胞株及胃癌组织中的表达情况。结果胃癌细胞系MGC-803癌细胞中girdin蛋白表达水平最高;girdin蛋白在胃癌组织中的表达率(54.1%)显著高于远癌胃黏膜组织中girdin蛋白的表达率(36.3%),差异有统计学意义(P<0.05);girdin蛋白的表达与胃癌患者Lauren分型,淋巴结转移和TNM分期显著相关(P均<0.05);girdin蛋白阳性表达组的胃癌患者生存时间显著短于阴性组的胃癌患者,预后较差(χ2=4.685,P<0.05);多因素COX回归分析显示girdin蛋白表达、组织学分型、远处转移和TNM分期是影响胃癌患者预后的独立危险因素。结论 girdin作为癌基因参与胃癌发生发展,girdin可能成为评估胃癌患者预后的生物标志。更多还原

【Abstract】 Objective To investigate the expression of girdin protein in gastric carcinoma and its correlation with clinical significance and prognosis of patients. Methods Immunohistochemical and Western blotting methods were used to detect the expression of girdin in gastric cancer cell lines and gastric carcinoma tissues. Results There was the highest level of girdin protein expression in gastric cancer cell line MGC-803. The expression of girdin protein was significantly higher in gastric carcinoma tissues(54. 1%) than in far from cancerous tissues(36. 3%),(P < 0. 05). The expression of girdin protein was significantly associated with Lauren’s classification,lymph node metastasis and the TNM staging of gastric cancer(P < 0. 05). The overall survival time of girdin protein positive expression group in gastric cancer patients was significantly shorter than that of the negative group(χ2= 4. 685,P < 0. 05). Multi-factors COX regression analysis results showed that girdin protein expression,lymph node metastasis,distant metastasis and TNM stage were independent risk factors affecting the prognosis of patients with gastric cancer. Conclusion Girdin,as cancer gene involved in gastric cancer development,may become the biomarker which can evaluate the prognosis of patients with gastric cancer.更多还原
