CD82抑制口腔鳞癌细胞OSCC-15裸鼠移植瘤生长的研究
Inhibitory effect of CD82 on xenograft in oral cancer cell OSCC-15 of nude mice
【摘要】 目的探讨CD82对口腔鳞癌细胞OSCC-15裸鼠移植瘤生长的影响。方法体外培养口腔鳞癌细胞株OSCC-15,将处于对数生长期的OSCC-15细胞接种于裸鼠右前肢皮下,建立裸鼠移植瘤模型。将荷瘤裸鼠随机分为空白对照组(人口腔鳞癌OSCC-15细胞)、阴性对照组(转染空白质粒载体的OSCC-15细胞)、实验组(转染pIRES2-EGFP-CD82过表达载体的OSCC-15细胞),每组6只。接种后25 d,处死裸鼠,称瘤体质量,测量肿瘤的最长径和最短径,计算肿瘤体积及体积抑制率。免疫组织化学法检测各组裸鼠移植瘤标本增殖细胞中的核抗原Ki67、增殖细胞核抗原(PCNA)表达水平。结果 CD82实验组移植瘤瘤体质量为(0.44±0.37)g,较空白组和阴性对照组小;体积抑制率为(77.21±3.25)%,较空白组和阴性对照组高,差异有统计学意义(P<0.05)。免疫组化结果显示,实验组Ki67,PCNA阳性染色细胞数分别为(348.51±117.09)个和(288.18±136.05)个;空白对照组为(522.23±101.17)个和(542.12±201.25)个;阴性对照组为(538.36±105.72)个和(558.33±205.22)个。实验组表达明显低于其他两组,差异有统计学意义(P<0.01)。结论 CD82对口腔鳞癌细胞裸鼠移植瘤具有一定的抑制作用。
【Abstract】 Objective To investigate the effect of CD82 on tumor growth in xenografted nude mice. Methods OSCC-15 cells in logarithmic phase were cultured in vitro and subcutaneously inoculated into the right foreleg of the nude mice. The mice were randomly divided into three groups( n = 6 in each group) : blank group( non-transfected cells),negative group( cells transfected with NC plasmid),and experimental group( cells transfected with CD82). The mice were sacrificed at 25 d after injection,and the tumors were removed and weighed. The tumor volume and inhibitory rate were calculated according to the longest and shortest size of tumors. The expression of Ki67,PCNA in specimen was detected by immunohistochemistry. Results The tumor weight in experimental group was( 0. 44 ± 0. 37) g,lighter than that of the other groups( P < 0. 05). The inhibitory rate in experimental group was( 77. 2 ± 3) %,higher than that of the other groups( P < 0. 05). The numbers of Ki67 and PCNA positive staining cells were 348. 51 ± 117. 09 and 288. 18 ±136. 05 in experimental group,522. 23 ± 101. 17 and 542. 12 ± 201. 25 in blank group,538. 36 ± 105. 72 and 558. 33 ± 205. 22 in negative group,respectively. The numbers of Ki67 and PCNA positive staining cells were significantly less in experimental group than in the other groups( P < 0. 01). Conclusion CD82 could inhibit oral squamous cell carcinoma cells growth in BALB/c nude mice.
