黑豆皮中花青素成分鉴定及细胞内抗氧化活性研究
Identification of the Anthocyanins Composition and Study on Cellular Antioxidant Activity from Black Soybean Coats
【摘要】 以黑豆皮为研究材料,分析鉴定黑豆皮中所含花青素的成分及组成,并对纯化所得黑豆皮花青素(Black soybean coats anthocyanins,BSCA)的体内外抗氧化活性进行研究。高效液相色谱(high performance liquid chromatography,HPLC)分析结果表明,BSCA的主要成分是矢车菊素-3-O-葡萄糖苷(C3G),占总样品含量的比例为64%,其含量为10.02 mg/g;所含其他成分分别是飞燕草素葡萄糖苷(D3G)占6.7%,矮牵牛素葡萄糖苷(Pt3G)占3.4%,矢车菊素占20.5%。体外化学方法检测结果表明,BSCA具有较强的DPPH与羟自由基的清除能力。细胞抗氧化活性(Cellular antioxidant activity,CAA)法研究表明,随着BSCA作用浓度的增加,Hep G2细胞内的荧光值降低,其抗氧化性增强,并且BSCA可不同程度的促进SOD,CAT,GSH-Px,GSH指标的升高和MDA含量的减少。MTT检测结果发现,BSCA对3种癌细胞都具有潜在的抗增殖作用,且呈一定的时间-剂量依赖性。可见黑豆皮花青素(BSCA)可作为抗氧化保健产品,对人体起到潜在的保护作用。
【Abstract】 Black soybean coats were used as research materials to analyze and identify the components of anthocyanins from black soybean coats. The antioxidant activity of black soybean coats anthocyanins(BSCA) was also studied in vitro and in vivo. High performance liquid chromatography(HPLC)analysis showed that the main component of BSCA was cyanidin-3-O-glucoside, whose proportion was 64% of the total sample, and purity was 10.02 mg/g. And the rest components were delphinidin-3-glucoside(6.7%),petunidin 3-glucoside(3.4%)and cyanidin(20.5%), respectively. The results of chemical detection in vitro showed that BSCA had strong DPPH and hydroxyl radical scavenging ability. The cellular antioxidant activity(CAA)assay showed that the fluorescence value of Hep G2 cells decreased, with the increase of BSCA concentration, and its antioxidant activity was enhanced. Furthermore, BSCA could induce to increase the activities of catalase(CAT), superoxide dismutase(SOD), glutathione(GSH) and glutathione Peroxidase(GSH-Px), whereas to reduce the contents of malonaldehyde(MDA)compared with the control cells. MTT assay showed that BSCA showed significant anti-proliferative effect on various cancer cells growth in a dose and time dependent manner. Therefore, BSCA may be regarded as an antioxidant health food to protect and benefit our health.
