紫外-可见分光光度法测定沙棘叶总黄酮含量的优化试验

【摘要】 笔者对紫外-可见分光光度法测定沙棘叶总黄酮含量的方法进行优化。以芦丁为标准品,Na NO2-Al(NO3)3-Na OH为显色体系,对参比空白、稀释定容乙醇的浓度、显色剂用量与显色时间进行条件试验。研究得出测定沙棘叶总黄酮的优化方法为:取1 m L样液于25 m L容量瓶中,加入5%Na NO20.3 m L,再加入10%Al(NO3)30.3 m L,摇匀静置6 min,然后加入4%Na OH 4 m L,摇匀静置10 min,用70%乙醇定容至刻度,以只加样品溶液为空白参比,于510 nm处测定吸光度值,计算沙棘叶总黄酮含量。该方法试剂用量少、时间短,可用于沙棘叶总黄酮含量的测定。

【Abstract】 Determination method of total flavonoids content in Hippophae rhamnoides leaves was studied by using the method of UV-visible spectrophotometry. The experiment was done on the factors such as blank reference,concentration of ethanol for diluting constant volume,chromogenic agent dosage and chromogenic time with rutin as standard material,Na NO2-Al( NO3)3-Na OH as chromogenic system. The results showed that the optimization methods were that took 1 m L sample solution in a 25 m L volumetric flsk,added 5% Na NO20. 3 m L and 10% Al( NO3)30. 3 m L,shaked well and reacted 6 min,then added 4% Na OH 4 m L,shaked well and reacted 10 min,absorbance was determined at 510 nm with sample as blank reference and total flavonoids content in Hippophae rhamnoides leaves was calculated. The optimized method with fewer reagent and shorter time could be used for the determinination of total flavonoids in Hippophae rhamnoides leaves.
