沙葱总酚含量及体外抗氧化活性与ACE酶抑制活性的研究
Total phenolic content, antioxidant activity and inhibitory effects against ACE enzyme of Allium mongolicum Regel
【摘要】 以沙葱脱脂干粉为原料制备水提物,测定其总酚含量和ACE酶的抑制活性,并且评价其体外抗氧化活性。采用Folin-Ciocalteu法测定沙葱水提物中总酚含量,利用清除2’-联氨-双-3-乙基苯并噻唑啉-6-磺酸[2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS]自由基抗氧化法,以及铁离子还原能力(Ferric-reducing antioxidant power,FRAP)法对其进行抗氧化活性评价,并且采用紫外法对ACE酶抑制活性进行测定。结果表明,沙葱水提物中总酚含量为10.20 mg GAE/g DW,ABTS法测定得到的Trolox等价抗氧化能力为9.55 mg Trolox/g DW,半抑制浓度为4.94 mg/mL,FRAP值为107.26μmol Fe2+/g,沙葱水提物对ACE酶活性抑制作用的IC50值为18.57 mg/mL。综上所述,沙葱是具有抗氧化活性与ACE酶抑制活性的天然植物酚类的潜在来源,可能在高血压防治方面发挥功效,可用于降压类功能性产品的开发。
【Abstract】 The aqueous extract of Allium mongolicum Regel(AM) was prepared to evaluate its total phenolic content, antioxidant capacity and in vitro inhibitory effects against ACE enzyme. The total phenolic content was determined by Folin-Ciocalteu assay and the antioxidant potential was estimatedby two methods: 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) method with results expressed as IC50 and TEAC, ferric reducing antioxidant potential assay(FRAP) method with results expressed as FRAP value. Additionally, the inhibitory activity of ACE enzyme was assessed by UV method. Results showed that the total phenolic content was 10.20 mg GAE/g DW, the TEAC value of ABTS radical scavenging activity of the extract was 9.55 mg Trolox/g DW and the IC50 value of scavenging abilities on ABTS was 4.94 mg/mL, the FRAP value was 107.26 μmol Fe2+/g. In addition, it also showed inhibitory activity against ACE enzyme in a certain extent with IC50 of 18.57 mg/mL. In summary, AM could be a potential source of natural plant phenolics which exhibit antioxidant activity and ACE enzyme inhibitory activity that may be useful in the prevention and control of hypertension. It suggests that AM could be used for the development of functional products of depressurization.
