葡萄皮多酚抗氧化性及提取工艺研究
Antioxidant activity and extract technology of polyphenols from grape skin
【摘要】 以酿酒葡萄皮为原料,首先探讨了不同溶剂和不同提取方式对葡萄皮提取物多酚得率和抗氧化性影响,然后研究了葡萄皮多酚的提取工艺。研究结果表明,采用甲醇、乙醇、丙酮和酸性水4种溶剂分别提取葡萄皮时,丙酮提取物多酚得率最高、总还原力最强、对DPPH·的清除作用最好;酸性水提取物对·OH清除效果最好。采用空气摇床、热回流、水浴振荡、静止浸渍4种方式分别提取葡萄皮时,采用空气摇床时的提取液的多酚含量最高、总还原力最强,清除DPPH·自由基能力最强;采用静止浸渍时提取液清除·OH自由基能力最强。采用热回流方式提取葡萄皮多酚的最优提取条件为:温度为66℃、提取时间为40 min、液料比为10、乙醇体积分数为52%,多酚理论提取得率可达到4.28%。
【Abstract】 Effects of different solvents and extracting methods on the total polyphenol content and antioxidant activity were studied for the grape skin. Then the extract technology was studied. Four kinds of solvents such as methanol, ethanol, acetone and acidic water were used to extract polyphenol from grape skin respectively. The acetone extract showed the highest polyphenol content, which has the highest total reducing power, the DPPH· radical and H2O2 scavenging capacity. The acidic water extract showed the highest ·OH radical scavenging capacity. Four kinds of extracting methods such as air incubator shakers extraction, heat reflux extraction, thermostatic water bath oscillators extraction and static immersion extraction were used to extract polyphenol from grape skin respectively. The extract with air incubator shakers showed the highest polyphenol content, which has the highest total reducing power, the DPPH· radical scavenging capacity. The extract with static immersion showed the highest ·OH radical scavenging capacity. The optimize conditions with heat reflux extraction method are extracting temperature 66 ℃, extracting time 40 min, liquid-to-solid 10 and ethanol concentration 52%. Under these conditions, the theoretical extracting yield of polyphenol is 4.28%.
