樟芝中4-乙酰基安卓奎诺尔B的提取方法筛选和优化

【摘要】 牛樟芝为原产于中国台湾地区的珍稀药用真菌,含有多种具有抗癌抗炎活性的物质,其中4-乙酰基安卓奎诺尔B已被证实具有良好抗癌活性,为泛锟类小分子物质。文章采用单因素优化法对牛樟芝中的4-乙酰基安卓奎诺尔B进行了提取工艺的优化研究。使用高效液相色谱法检测4-乙酰基安卓奎诺尔B的含量。结果表明,牛樟芝中4-乙酰基安卓奎诺尔B的最优提取方法为振荡提取法;最优提取条件如下:温度为30℃,转速为150 r/min,溶剂为90%的乙醇,料液比为1:20(g/m L),时间为1.5 h。在此提取条件下,牛樟芝中4-乙酰基安卓奎诺尔B提取率达到97%以上,提取一次即可。该方法操作简单、节约溶剂、缩短时间。

【Abstract】 This paper discussed the extraction process of 4-acetylantroquinonol B which was identified to be the major bioactive compound of Antrodia cinnamomea cultivated by submerged fermentation. This experiment was based on the single factor method. The results showed that the optimal extraction method for 4-acetylantroquinonol B in Antrodia cinnamomea was by oscillatory extraction. The experimental results showed that the optimal conditions for the extraction were as follows: material ratio 1:20(g/m L), the extraction solvent was 90% ethanol, the extraction speed was 150 r/min, the extraction temperature was 30 ℃, the extraction time was 1.5 h. In such a condition of extraction, the extraction rate of 4-acetylantroquinonol B in Antrodia cinnamomea can reach more than 97% at one time. The method is simplicity of operator and could save solvent.
