青钱柳抑制α-淀粉酶活性筛选研究

【摘要】 目的:研究青钱柳提取物对α-淀粉酶的抑制作用。方法:采用不同溶剂(石油醚、二氯甲烷、乙酸乙酯、丙酮、无水乙醇和水)回流提取和不同溶剂(石油醚、乙酸乙酯、正丁醇)萃取法制备青钱柳提取物,采用α-淀粉酶的抑制模型评价其活性筛选最佳提取方法;在此基础上,研究不同来源青钱柳对α-淀粉酶的抑制活性。结果:石油醚回流提取法获得的提取物对α-淀粉酶活性抑制最好;在30 mg/m L时,不同来源青钱柳石油醚提取物的抑制率分别为:江西修水(36.4%),湖南八大公山(71.0%),湖南张家界(67.6%),湖南绥宁(58.3%),湖南邵阳(59.6%),贵州榕江平阳(35.3%),贵州榕江小丹江(65.7%),贵州贵阳(67.7%)。结论:石油醚回流提取法适于获得青钱柳α-淀粉酶抑制活性成分;不同来源青钱柳提取物对α-淀粉酶均具有抑制作用,但存在差异性。

【Abstract】 Objective: The inhibitory effect of Cyclocarya paliurus extracts on α-amylase activity were investigated. Method: The extracts were refluxed with different polar solvent, namely petroleum ether, dichloromethane, ethyl acetate, acetone, ethyl alcohol and water, while with petroleum ether, ethyl acetate, n-butyl alcohol extracting orderly. The inhibitory effect of different extracts were evaluated by α-amylase. The best extraction method and effective fraction were screened, and these method is also used to obtain the effective parts of Cyclocarya paliurus from different sources and to evaluate their inhibitory activity of α-amylase. Result: The extracts obtained by reflux with petroleum was the best effective fraction. At 30 mg/m L, the inhibition rates of petroleum ether extract of Cyclocarya paliurus from different sources were as follows: Xiushui Jiangxi(36.4%), Badagongshan Hunan(71.0%), Zhangjiajie Hunan(67.6%), Suining Hunan(58.3%), Shaoyang Hunan(59.6%), Pingyang Rongjiang Guizhou(35.3%), Xiaodanjiang Rongjiang Guizhou(65.7%) and Guiyang Guizhou(67.7%). Conclusion: The extraction of petroleum by reflux was suitable for obtaining the α-amylase inhibitory activity components. The extracts from different sources had inhibitory activity on α-amylase, but their activities had differences.
