超高效液相色谱质谱联用仪在发酵豆乳中异黄酮分析的应用
The application of UPLC-MS on analysis of isoflavone in fermented soybean milk
【摘要】 目的:建立同时测定发酵豆乳中大豆苷、大豆苷元、染料木苷的超高效液相色谱质谱方法。方法:采用Acquity UPLC BEH C18(1.7μm,2.1 mm×100 mm)色谱柱进行分离,0.1%甲酸水溶液(A)与0.1%甲酸乙腈溶液(B)作为流动相,并且进行梯度洗脱。色谱柱温度35℃,进样体积5μL。质谱采用电喷雾离子源正离子模式进行采集,离子源温度100℃,脱溶剂气温度450℃,脱溶剂气流量600 L/h。结果:通过绘制大豆苷、大豆苷元、染料木苷的标准曲线,得到其线性范围分别是(4.00100)mg/L、(0.04100)mg/L、(0.04100)mg/L,最低检出限分别是5.00、0.85、1.00μg/L,最低定量限分别是15.0、2.55、3.00μg/L,相关性系数范围为0.9910.993,平均回收率范围89.50%107.33%。研究为发酵豆乳中异黄酮的检测提供了高效、准确的方法。
【Abstract】 Objective: To develop a method to simultaneously determine the daidzin, daidzein and genistein in fermented soybean milk by using UPLC-MS. Methods: The samples were separated on Acquity UPLC BEH C18(1.7 μm, 2.1 mm×100 mm) column with mobile phase of 0.1% formic acid aqueous solution(A) and 0.1% formic acid acetonitrile solution(B) in gradient mode. The column temperature was set at 35 ℃, and the injection volume was 5 μL. The ESI positive mode was selected to monitor the changes, and the MS parameters were as follows: source temperature 100 ℃, desolvation gas temperature 450 ℃, desolvation gas flow 600 L/h. Results: By drawing the curve of daidzin, daidzein and genistein, the linearity range were as follows:(4.00100)mg/L,(0.04100)mg/L,(0.04100)mg/L respectively; the minimum detection limits were 5.00, 0.85, 1.00 μg/L respectively; the minimum quantitation limits were 15.0, 2.55, 3.00 μg/L respectively; the range of correlation coefficient R2 was between 0.991 to 0.993; and the recoveries varied between 89.50% to 107.3%. This method is efficient and accurate, and provides reliable basis for future study on the quality of fermented soybean milk.
