HPLC-MS法测定罗汉果中6种罗汉果甜苷含量
Determination of Content of 6 Kinds of Mogrosides in Siraitia grosvenorii by Using HPLC-MS/MS
【摘要】 利用高效液相色谱串联质谱(high performance liquid chromatography-mass spectrometry,HPLC-MS),分析比较不同产地罗汉果中6种罗汉果甜苷(11-氧-罗汉果皂苷V、罗汉果皂苷V、罗汉果皂苷IV、赛门苷I、罗汉果皂苷III E和罗汉果皂苷II E)的含量,同时利用该方法分析罗汉果甜苷提取和大孔树脂D101纯化过程中该6种成分的含量变化情况。样品经甲醇超声提取,色谱分离采用ZORBAX SB-Aq色谱柱(4.6 mm×150 mm,5μm),梯度进行洗脱,柱温25℃,流速0.5 m L/min。质谱分析采用电喷雾电离源(electrospray ionization,ESI),正离子模式检测。结果表明:6种甜苷在0.75 mg/L~9.00 mg/L范围内呈现良好的线性关系(r>0.9975),检测限为0.38 mg/kg~1.28 mg/kg,平均加样回收率89.3%~105.6%。经分析测定发现,桂林市永福百寿乡适宜种植高品质罗汉果,利用大孔树脂D101可很好地进行罗汉果甜苷富集和纯化,同时利用的HPLC-MS方法,可为罗汉果种植和生产提供质量分析控制方法。
【Abstract】 A method for the determination of six mogrosides(11-oxo-mogroside-V,mogroside-V,mogrosideIV,siamenoside-I,mogroside-IIIE and mogroside-IIE)in Siraitia grosvenorii was established and validated by(high performance liquid chromatography-mass spectrometry,HPLC-MS). And this method was used to analyze the concentrations of the 6 components of Siraitia grosvenorii of different habitats and the products from the process of the purification of sweet glycosides of Siraitia grosvenorii. The samples were extracted with methanol by using ultrasonic. The extracts were separated by ZORBAX SB-Aq column(4.6 mm× 150 mm,5 μm)with gradient elution,and temperature was 25 ℃,and speed was 0.5 m L/min. The mass spectrometric conditions were electrospray ionization(ESI),positive ion mode. The results showed that this six compounds had good linear relationship(r > 0.997 5). In the range of the detection limits were 0.75 mg/L-9.00 mg/L,the detection limits were 0.38 mg/kg-1.28 mg/kg,the average recoveries were 89.3 %-105.6 %. The analysis showed that Yongfu county Baishou town is suitable for planting high quality Siraitia grosvenorii,using macroporous resin D101 can easily enrich and purify the sweet glycosides of Siraitia grosvenorii. In additional,using the method of HPLCMS can provide methodological support for the establishment of the cultivation and production quality standards of Siraitia grosvenorii.
