低温淀粉酶菌种分离纯化与发酵生产研究
Isolation Purification and Fermentation Production of Low Temperature Amylase
【摘要】 通过对滨州黄河三角洲地区的样品中微生物的采集,经过选择性培养基分离获得了4株产葡萄糖淀粉酶较高的菌株:Bz11、Bz25、Bz38、Bz73,其在20℃可以产生较高活性的淀粉酶,与报道菌株降低10℃左右,经过复筛最终选择产酶相对较高的Bz38作为研究对象,通过平板菌落观察、显微镜细胞观察,碳源发酵与同化试验、26Sr DNA分子生物学鉴定,最终确定Bz38为解脂亚罗酵母菌(Yarrowia lipolytica)。在掌握菌株生长规律的基础上,选择碳源(葡萄糖、蔗糖、乳糖、麦芽糖、甘露醇)、氮源(牛肉膏、酵母膏、蛋白胨、尿素、酪蛋白胨、硫酸铵)单因子、菌株淀粉酶活达到960 U/m L,为全面应用葡萄糖淀粉酶奠定了坚实的力量基础,也极大的丰富了本地区对微生物种质资源的开发和利用。
【Abstract】 Through the collection and separation of microorganisms on Binzhou the Yellow River delta samples,obtaining four strains producing high glucose starch:Bz11,Bz25,Bz38,Bz73,it could produce high active amylase at 20 ℃,and decreasing about 10 centigrade with the reported strain. the final choice of enzyme production of relatively high Bz38 was as the research object,observation the colony on plate culture,cell observation by microscope,experiment of carbon source fermentation and assimilation,identification of26 Sr DNA molecular biology,Bz38 was eventually identified as Yarrowia lipolytica. On the basis of mastering the law of growth of Bz38.Single factor carbon(glucose,sucrose,lactose,maltose,mannitol)and nitrogen source(beef paste,yeast extract,peptone,urea,casein peptone,ammonium sulfate)experiments were carried out,The amylase activity of strain reached 960 U/m L,for the full application of glucose amylase laid a solid foundation for strength,it also greatly enriched the development and utilization of microbial planting resources in this region.
