人不同部位冻存颗粒脂肪活性的实验研究

【摘要】 目的探索不同部位冻存颗粒脂肪活性的差异,为临床脂肪移植供区选择提供理论依据。方法自2016年6~10月,常规收集45例行大腿内侧、大腿外侧、腹部吸脂术患者术后的颗粒脂肪。将标本添加等量冻存保护液后,于-80℃深低温冰箱中冻存6个月;之后将标本复苏,用HE染色并观察组织形态,计算完整细胞比率;用锥虫蓝染色,计算脂肪细胞拒染率;通过肌酸激酶实验来测定标本活性。结果 HE染色:大腿内侧脂肪细胞完整率为(78.30±2.03)%;大腿外侧为(70.50±4.53)%;腹部为(65.10±4.63)%。各组之间差异有统计学意义(P<0.05)。锥虫蓝染色、肌酸激酶结果与HE染色一致。结论冻存6个月后,大腿内侧颗粒脂肪活性高于大腿外侧及腹部。

【Abstract】 Objective To explore the viability of cryopreserved fat granules from different parts of the human body and to provide a theoretical basis for donor site selection in clinical adipose tissue transplantation. Methods From June to October 2016, fat granules from the femoribus internus, lateral thighs, and abdomen of 45 conventional liposuction patients were collected. All samples were stored in-80℃ refrigerators after adding equal amounts of cryopreservation solution. Six months later, the specimens were rewarmed. H&E staining was used to observe the morphology, and the ratio of intact adipocytes was calculated. The percentage of dead adipocytes was calculated with trypan blue staining. The vitality of fat granules was measured through creatine kinase experiment. Results H&E staining showed that the rate of intact adipocytes in the femoribus internus was(78.30±2.03)%, in the lateral thigh was(70.50±4.53)%, and in the abdomen was(65.10±4.63)%. The differences among the three groups were statistically significant(P<0.05). The results of trypan blue staining and creatine kinase experiment were consistent with the H&E staining. Conclusion After 6 months of cryopreservation, the viability of fat granules in the femoribus internus was higher than that in the lateral thigh and abdomen.
