酿酒酵母模型重金属镉毒性拮抗药物筛选
Screening the Antagonistic Drug of the Heavy Metal Cadmium Toxicity in Saccharomyces cerevisiae Model
【摘要】 重金属镉极易在人体内蓄积且自然排泄缓慢,对人体健康危害严重,临床上目前还没有有效治疗镉中毒的特异性药物,因此镉毒性的干预措施及机理研究意义重大。实验以酿酒酵母为模型,研究了在20μmol/L镉暴露条件下,加入Zn、Mg、Se、谷胱甘肽、维生素C和部分氨基酸组成的混合物对镉毒性的拮抗作用。单因素实验结果显示,所选择的微量元素,维生素和氨基酸混合物都能起到缓解镉暴露引起酵母生长的抑制作用,但达不到完全恢复酵母生长的效果。采用同时添加35μmol/L的Zn,30μmol/L的维生素C,5μmol/L的镁、硒和谷胱甘肽以及5mL/L氨基酸混合物,则能完全恢复镉抑制酵母的生长。进一步研究显示,混合物能有效阻止镉在酵母细胞中的蓄积。
【Abstract】 Cadmium(Cd) is easily accumulated in humans but difficult to be discharged by organism itself. Therefore, the accumulated Cd causes serious effects on human health. Since no effective therapy for Cd poisoning is available, it is very important to explore for the methods and mechanisms of antagonistic Cd toxicity. In this paper,the inhibiting Cd toxicity by nutrient elements(zinc, magnesium, selenium and vitamin C), glutathione(GSH)and mixture of amino acid were investigated in saccharomyces cerevisiae model. The result showed that supplement of the selected trace elements, vitamins and amino acids could reduce the inhibitory growth of yeast exposure to 20μmol/L of cadmium, but dose not completely restore. However, the growth was restored completely by adding 35μmol/L of Zn, 30μmol/L of vitamin C, 5μmol mol/L of magnesium, selenium and glutathione, and 5 mL/L amino acid mixture. The mixture antagonism could effectively prevent the accumulation of cadmium in yeast cells.
