去势对雄性大鼠海马组织中TMP-21、IDE、NEP表达的影响
Effects of castration on the expression levels of TMP-21,IDE and NEP in the hippocampus of male rat
【摘要】 目的:观察去势对雄性SD大鼠海马组织中转运膜蛋白21(transmembrane protein 21,TMP-21)、胰岛素降解酶(insulin-degrading enzyme,IDE)、脑啡肽酶(neprilysin,NEP)的表达影响,进一步探讨雄激素对TMP-21、IDE、NEP表达的调节作用。方法:2月龄雄性SD大鼠随机分为假手术组(Sham)和去势手术组(GDX),术后30 d断头取脑剥离海马组织,一侧用于RT-PCR的方法检测TMP-21、IDE、NEP的mRNA水平,另一侧用于Western Blot的方法检测TMP-21、IDE、NEP的蛋白水平。结果:与Sham组相比,海马组织中GDX组TMP-21、IDE、NEP的mRNA水平分别下降了约28%、63%、44%(P<0.05),蛋白水平分别下降了约39%、48%、26%(P<0.05)。结论:去势可引起雄性SD大鼠海马组织中TMP-21、IDE、NEP mRNA水平和蛋白水平的显著降低,提示TMP-21、IDE、NEP表达水平的降低可能与雄激素的减少相关。
【Abstract】 Objective: To observe the effects of castration on the expression levels of transfer membrane protein 21( TMP-21),insulin degradation enzyme( IDE),enkephalin enzyme( NEP) in male SD rat hippocampus,and further to discuss the expression of androgens for TMP-21,IDE,NEP. Methods: Male 2-month-old Sprague-Dawley rats were randomly divided into sham-gonadectomized group( sham group) and gonadectomized group( GDX group). After 30 days,all amimals were killed and the hippocampus were dissected,one side was used to detect the mRNA levels of TMP-21,NEP,IDE with RT-PCR,the other side was used to detect the protein levels of TMP-21,IDE,NEP with the method of Western Blot. Results: Compared with Sham group,the mRNA levels of TMP-21,IDE,NEP in GDX group were respectively decreased by 28%,63% and 44%( P < 0. 05),and the protein levels decreased by about 39%,48% and 26%( P < 0. 05). Conclusion: Castration could significantly reduce the mRNA and protein levels of TMP-21,IDE,NEP in male SD rats hippocampus,indicated that the low expression level of TMP-21,IDE,NEP may be associated with the decrease of androgen.
