抵当汤对颈动脉损伤后再狭窄大鼠VEGF-A、AngⅡ的影响

【摘要】 目的:观察抵当汤对实验性颈动脉损伤后再狭窄大鼠VEGF-A、AngⅡ水平的影响。方法:50只SD雄性大鼠随机分为正常对照组、模型组、中药大剂量组、中药小剂量组和西药组,每组10只。用自制导管对大鼠右颈总动脉进行损伤,造成颈动脉狭窄模型。术前2 d开始给药,中药大、小剂量组以抵当汤(2.4 g/kg、1.2 g/kg生药剂量)、西药组以肠溶阿司匹林30 mg/kg灌胃,模型组和正常对照组以等容积生理盐水灌胃。术后2 w,腹主动脉取血,离心后酶联免疫吸附法(ELISA)检测血浆VEGF-A、AngⅡ水平,生化法检测血脂含量。结果:与正常对照组比较,模型组VEGF-A、AngⅡ水平显著升高(P<0.01);与模型组比较,抵当汤大、小剂量组和西药组VEGF-A、AngⅡ水平均显著降低(P<0.01);抵当汤小剂量组VEGF-A、AngⅡ水平显著低于大剂量组和西药组(P<0.01,P<0.05);与抵当汤大剂量组比较,西药组VEGF-A显著降低(P<0.05)。抵当汤大、小剂量组和西药组TC、TG、LDL-C含量均低于模型组(P<0.01),HDL-C含量均高于模型组(P<0.01)。结论:抵当汤可有效抑制实验性颈动脉损伤后再狭窄大鼠VEGF-A、AngⅡ水平,其作用以低剂量为佳。

【Abstract】 Objective:To observe the effect of Didangtang on contents of plasma VEGF-A and AngⅡ of rat models with restenosis after carotid artery injury. Methods:SD male rats were randomly divided into normal group,model group,traditional Chinese medicine high dosage group,low dose group and Western medicine group,10 rats in each group. The injury in right common carotid artery of rats was induced by a self-made catheter. High and low dosage groups of Didangtang were given respectively 2.4 g/kg,1.2 g/kg crude drug dosage and Western medicine group was given aspirin 30 mg/kg,while the normal group and model group were given the same volume of saline 2 days before operation. Blood samples were taken from abdominal aorta,then contents of plasma VEGF-A、Ang Ⅱ were detected by enzyme linked immunosorbent assay(ELISA)2 weeks after operation. Results:Compared with the normal group,contents of VEGF-A and AngⅡ in model group were increased significantly(P<0.01);compared with the model group,contents of VEGF-A and AngⅡ in treatment groups were significantly decreased(P<0.01);contents of VEGF-A and AngⅡ in small dosage Didangtang group was significantly lower than those in other treatment groups(P<0.01,P<0.05);compared the Didangtang high dosage group,VEGF-A content in Western medicine group was decreased significantly(P<0.05). Levels of TC,TG and LDL-C in treatment groups were lower than those in model group(P<0.01),HDL-C level was higher than that in model group(P<0.01).Conclusion:Didangtang could effectively inhibit the contencs of VECF-A and Ang Ⅱ of experimental restenosis after carotid artery injury,and Didangtang of low dosage has better effect.
