口腔菌属特异性16S rRNA基因序列V1区的特征分析

【摘要】 为了确定针对属特异性16S rRNA基因序列V1区设计的引物位点的特异性.基于人类口腔微生物菌群16S rRNA的基因库,对其进行属水平上序列的特征分析,得出其序列突变图谱,找出V1区引物设计位点,计算该引物位点在其分属中的覆盖率并与其通用引物在各分属中的覆盖率进行比较.结果表明,17个菌属中有13个菌属的V1区引物位点在菌属中的覆盖率接近或超过90%,而4个通用引物在菌属中呈现低的覆盖率.由此得出结论,针对属特异性设计的引物位点比通用引物在各菌属中有着更好的覆盖率.该技术对于口腔和食品、肠道、环境等微生物菌群的菌种鉴定和评估方面有着重要的意义.

【Abstract】 This study is to determine the specificity of the genus-specific primers designed according to V1 region of16 S rRNA gene for each genus. The genus-level specificity of 16 S rRNA gene sequence was analyzed and the mutations map for each genus was made,based on human oral microbiota in the 16 S rRNA gene library. The primer sites were found for V1 region,and the coverage of the primer sites was calculated in the genus and compared to the universal primers coverage for each V1 region. The results showed that the coverage of the V1 region of 13 out of the 17 genera was close to or exceeded 90% while the four universal primers showed low coverage in the genus. It is therefore concluded that primer-specific primers have better coverage in each genus than universal primers. This technique is of great significance to the identification and evaluation of microbial flora in the oral cavity,food,gut and environment,etc.
