明目消朦预处理对H2O2诱导人视网膜色素上皮细胞凋亡及氧化应激损伤的影响

【摘要】 目的观察明目消朦(MMXM)预处理对H2O2诱导人视网膜色素上皮细胞(RPE细胞)凋亡和氧化应激损伤的影响。方法取对数生长的RPE细胞,将细胞分为4组,A组加入含MMXM的无血清培养基,MMXM的终浓度为0. 5 mg/m L,24 h后加入200μmol/m L的H2O2(以制备氧化应激损伤模型); B组加入无血清的基础培养基,24 h后加入200μmol/m L的H2O2; C组加入含MMXM的无血清培养基,MMXM的终浓度为0. 5 mg/m L; D组加入无血清的基础培养基。比较各组细胞调亡率及ROS、MDA、SOD、GSH-PX。结果与B组比较,其余3组细胞凋亡率降低(P均<0. 05);与C、D组比较,A、B组细胞凋亡率升高(P均<0. 05)。与D组比较,A、B组ROS(+)、MDA升高,ROS(-)、SOD、GSH-PX降低(P均<0. 05);与B组比较,其余3组ROS(-)、SOD、GSH-PX升高,ROS(+)、MDA降低(P均<0. 05)。结论 MMXM预处理能有效减少H2O2诱导的RPE细胞凋亡,并可预防细胞氧化损伤。

【Abstract】 Objective To observe the effects of pretreatment of Mingmu Xiaomeng( MMXM) on H2O2-induced apoptosis and oxidative stress injury in human retinal pigment epithelial cells( RPE cells). Methods RPE cells in the logarithmic growth were divided into 4 groups: groups A,B,C,and D. Cells in the group A were added with serum-free medium containing 0. 5 mg/m L MMXM,and 200 μmol/m L H2O2 was added after 24 h( to prepare an oxidative stress damage model); serum-free basal medium was added to the cells in the group B,and 200 μmol/m L H2O2 was added after 24 h;cells in the group C were added with serum-free medium containing MMXM with the final concentration of 0. 5 mg/m L; serum-free basal medium was added to the cells in the group D. The apoptosis rate and reactive oxygen species( ROS),malondialdehyde( MDA),superoxide dismutase( SOD) and glutathione peroxidase( GSH-PX) were analyzed. Results Group B had a higher apoptosis rate than the other three groups( P < 0. 05); compared with group C and D,the apoptosis rate increased in the group A and group B( P < 0. 05); compared with group D,ROS( +) and MDA increased,and ROS(-),SOD,and GSH-PX decreased in the group A and group B( all P < 0. 05); compared with group B,ROS(-),SOD,and GSH-PX increased and ROS( +) and MDA decreased in the other three groups( all P < 0. 05). Conclusion MMXM pretreatment can effectively reduce H2O2-induced apoptosis and prevent oxidative damage in RPE cells.
