HPLC法测定丁白贴剂成分含量的研究

【摘要】 建立了采用高效液相色谱法测定丁白贴剂中丁香酚、芍药苷含量的方法,采用Lichrospher C18柱(4.6 mm×150 mm,5μm)色谱柱,以甲醇-水(52:48,V/V)为流动相,流速1 mL·min-1,检测波长280 nm,柱温30℃,测定制剂中丁香酚的含量;以乙腈-水(13:87,V/V)为流动相,流速1 mL·min-1,检测波长230 nm,柱温30℃,测定制剂中芍药苷的含量。结果显示丁香酚进样量在0.047 20.472μg范围线性关系良好,R=1.000 0。芍药苷进样量在0.054 60.491 4μg范围线性关系良好,R=0.999 9。该方法简便、重复性好、准确可靠,可作为丁白贴剂的含量测定方法。

【Abstract】 A method was established to determine the content of Eugenol and paeoniflorin in Dingbai plaster by HPLC. The analysis was performed on Lichrospher C18(4.6 mm× 150 mm,5 μm) column,with the mobile phase of methanol-water(52: 48,V/V). At a flow rate of 1 mL·min-1,the detection wavelength was set at 280 nm. The column temperature was kept at 30 ℃,and the content of eugenol in preparation was determined; with the mobile phase of acetonitrile-water(13: 87,V/V),at a flow rate of 1 mL·min-1,the detection wavelength was set at 230 nm,the column temperature was kept at 30 ℃,and the content of paeoniflorin in preparation was determined. Better linear relationship exists in the range 0.047 2~0.472 μg(R= 1.000 0) for eugenol. And in the range 0.054 6~0.491 4 μg(R= 0.999 9) for paeoniflorin. The method is simple,reproducible,accurate and reliable. It can be used as a method for the determination of the content of Dingbai plaster.
