正交试验优化沙地油栗硒多糖提取工艺研究

【摘要】 选用世界硒都的恩施市沙地乡富硒板栗为原料,采用超声辅助水浴浸提方法提取其硒多糖,在分析单因素试验的基础上进行正交试验,优化了板栗硒多糖的提取工艺,并以硫酸-苯酚法测定硒多糖的含量,以原子荧光法检测硒多糖的含硒量。结果表明,板栗硒多糖最佳的提取条件为超声15min、温度60℃、料液比1∶15、浸提时间为1 h,在最佳的提取工艺下,板栗多糖的提取率为1.35%,并且测得板栗样品的硒含量为2.715μg/g,提纯多糖硒含量为0.429μg/g。

【Abstract】 The Selenium-rich chestnut in Shadi Township of Enshi City,was used as the raw material to get Se-polysaccharides from it by the water extraction method that combined with the ultrasonic procesing method. Based on the analysis of the single factor experiment,an orthogonal experiment was conducted to optimize the Se-polysaccharides extraction craft. The phenol sulfuric acid method was used to detect the content of Se-polysaccharide in the chestnut,and the atomic fluorescence method was used to detect the content of Se in the Se-polysaccharide. The results show that the best condition to extract the Se-polysaccharide were as follows: processing time of ultrasonic was 15 minutes,the solid-liquid ratio was 1 ∶ 15,water extraction time was 1,temperature was 60 ℃. Under this condition,selenium content of chestnut was 2. 715%,selenium content in the Se-polysaccharide was 0. 429μg/g.
