【摘要】 目的分析人外周血淋巴细胞染色体制备中的影响因素,做好质量控制,掌握染色体制备技术,及时准确反映辐射损伤情况,为诊断与治疗提供依据。方法采用微量全血培养法制备标本,以RPMI1640全培养基进行体外淋巴细胞培养,对采血后细胞培养时间、秋水仙素浓度、滴片等影响因素进行分析。结果 1 089例放射工作人员外周血淋巴细胞染色体标本中,1次培养成功1 077例,出现问题后采取补救措施补救成功12例,成功率100%。结论影响人外周血淋巴细胞染色体制备的因素很多,总结一套适合本实验室的制备方法 ,确保外周血染色体制备成功。更多还原

【Abstract】 Objective To analyze the influencing factors over the preparation of chromosomes in peripheral blood lymphocyte, to investigate how to improve quality control over the analysis of the chromosome aberration in peripheral blood lymphocyte culture and master the preparation technic of chromosomes, to reflect the radiation damage timely and accurately, to provide a basis for diagnosis and treatment of radiation damage. Methods Adopt method of micro-whole blood culture for the preparation of specimens. Lymphocytes were cultured in vitro with RPMI1640 complete medium. Influencing factors such as cell culture time, concentration of colchicines and droppings after blood collection were analyzed. Results Among 1089 cases of the chromosome specimens from peripheral blood lymphocyte of radiation workers, 1077 cases were cultured successfully at the first time operation and 12 cases were successfully remedied by taking remedial measures after problems occuring. The success rate is 100%. Conclusion The preparation of chromosomes in human peripheral blood lymphocytes could be affected by a number of factors. A set of operation method suitable for this laboratory should be summarized to ensure the successful preparation of chromosomes in peripheral blood.更多还原
