异育银鲫中鲤疱疹病毒Ⅱ型载量测定方法的建立
Establishment of the method to quantify the cyprinid Herpes virus Ⅱ in Carassius auratus gibelio
【摘要】 异育银鲫是我国重要的淡水养殖品种,但是鲤疱疹病毒Ⅱ型对其造成了重大经济损失,该病毒致死率几乎可以达到100%。为了能够有效预防和控制该病毒,需要建立一种高效灵敏的定量方法。为此,本实验室根据CyHV-16基因序列设计引物,利用SYBR Green法建立了异育银鲫中鲤疱疹病毒Ⅱ型Realtime PCR方法,可以定量鲫鱼体内疱疹病毒载量,绘制了标准曲线,根据各个组织中病毒的拷贝数分析出该病毒在不同组织中的表达分布,其中脾、肾组织中表达量较高,鳃、肝、肠、肌肉等组织中表达量较低。这些数据为预防和控制异育银鲫Ⅱ型疱疹病毒奠定了实验和研究基础。
【Abstract】 C. auratus gibelio is one of important freshwater aquaculture species in China. Cyprinid herpes virus Ⅱ can cause 100% fatality rate and great economic losses. In order to effectively prevent and control this disease, it is necessary to establish a sensitive and efficient quantitative method for this virus. So base on SYBR Green real-time PCR to quantify the Cyprinid herpes virus Ⅱ in the diseased crucian carp, primers of Cy HV-16 gene sequences were designed. The standard curve was structured firstly. Based on this data, the number of copies of virus in each tissue of diseased crucian carp were calculated. The results showed that the expression levels in the spleen and kidney were higher compared with the gill, liver, intestines, muscles and other tissues.These data laid the foundation for future research, which is beneficial to the control and prevention of Cyprinid herpes virusⅡ.
