糖氧剥夺/再灌注抑制L2.3神经干细胞N-Myc-DLL3-Notch信号系统
Glucose Deprivation/Reperfusion Inhibits N-Myc-DLL3-Notch Signaling System of L2.3 Neural Stemcells
【摘要】 目的建立L2.3神经干细胞糖氧剥夺/再灌注损伤模型,探讨糖氧剥夺/再灌注是否可以抑制L2.3神经干细胞N-Myc-DLL3-Notch信号系统。方法体外培养的L2.3神经干细胞经历糖氧剥夺(OGD)6h,再灌注(R)16h,建立OGD/R模型。分别在OGD前、OGD6h、OGD6h再灌注16h,用western blot方法检测Notch1、NICD、DLL3和N-Myc蛋白表达情况。结果与糖氧剥夺/再灌注前比,糖氧剥夺6h,再灌注16h可下调Notch1、NICD、DLL3和N-Myc蛋白表达。结论糖氧剥夺/再灌注损伤可抑制L2.3神经干细胞N-Myc-DLL3-Notch信号系统
【Abstract】 Objective To establish L2.3 neural stem cells model of glucose and oxygen deprivation/reperfusion injury,and to investigate whether glucose deprivation/reperfusion can inhibit N-Myc-DLL3-Notch signaling system of L2.3 neural stem cells.Methods L2. 3 neural stem cells in vitro cultured were given oxygen glucose deprivation( OGD) 6h and reperfusion( R) 16 h,and then a OGD/R model was established.The expression of Notch1、NICD、DLL3 and N-Myc protein was detected by western blot method at before OGD,OGD6h、16h after reperfusion,respectively.Results Compared with before oxygen and glucose deprivation/reperfusion,the expression of Notch1、NICD、DLL3 and N-Myc protein decreased after 6h of glucose and oxygen and glucose deprivation and 16h of reperfusion. Conclusion Oxygen and glucose deprivation/reperfusion injury can inhibit N-MycDLL3-Notch signaling system of L2.3 neural stem cells.
