菠萝对乙烯利诱花的敏感性差异研究

【摘要】 为了解造成2种不同类型菠萝品种对乙烯敏感性差异的发生环节,进一步揭示乙烯促进菠萝成花的分子机制,本研究以乙烯敏感型‘巴厘’菠萝品种和乙烯钝感型‘台农16’菠萝品种为试材,对乙烯催花过程中成花基因及乙烯信号转导途径中乙烯受体、信号转导和核内调控等过程涉及基因的表达特征进行了分析。结果表明:Ac ERS1a、Ac ETR2a、Ac ETR2b、Ac CTR1、Ac ENI2和Ac ERFs等基因在2种成花类型中的差异可能是导致2个品种对乙烯信号感知敏感度不同的重要原因;Ac ERS1a、Ac ERS1b、Ac ETR2a和Ac ETR2b等乙烯受体可能参与乙烯诱导菠萝成花信号的感知,然后通过Ac CTR1下调和Ac ENI2上调将信号转导至核内调控基因(Ac EIN3和Ac ERFs),从而启动菠萝成花关键基因Ac FT和花器官形态建成重要基因Ac AP1的表达,引起菠萝成花的生物学效应,这将为进一步阐明乙烯诱导菠萝成花的分子机制提供参考,也为利用基因工程手段开展菠萝新品种选育奠定了基础。

【Abstract】 In order to explore the sensitivity difference to ethylene between ethylene sensitive cultivar ‘Bali’ and ethylene insensitive cultivar ‘Tainong 16’, and further clarify the molecular mechanisms of flowering induced by ethylene in pineapple, the expression profiles of flowering genes and ethylene-related genes during floral induction were studied by the real-time quantitative PCR method. Results showed that the expression difference of Ac ERS1 a, Ac ETR2 a, Ac ETR2 b, Ac CTR1, Ac ENI2 and Ac ERFs during flower-bud formation might be the main reasons causing the difference in ethylene sensitivity of the two cultivars. The ethylene receptors Ac ERS1 a, Ac ERS1 b, Ac ETR2 a and Ac ETR2 b might participate in signal perception of flowering induced by ethylene, then by down-regulation of Ac CTR1 and up-regulation of Ac ENI2, the signal was transmited to Ac EIN3 and Ac ERFs, to initiate the expression of flowering genes Ac FT and Ac AP1, and raise the biological effects of floweing. The study would provide a reference to clarify the molecular mechanism of pineapple flowering induced by ethylene, and provide scientific bases for genetic engineering breeding.
