肺结核患者外周血T淋巴细胞上OX40的表达水平
OX40 expression in peripheral blood T lymphocytes in pulmonary tuberculosis patients
【摘要】 目的比较肺结核病人和健康人群外周血单个核细胞(PBMC)和T淋巴细胞上共刺激分子OX40的表达水平,以及辅助性T细胞亚群,包括Th1、Th2、Th9和调节性T细胞(Treg)在两组人群中的差异,进而探索OX40的表达水平与结核病进程以及CD4~+T细胞功能亚群分化的相关性。方法选取2015年11月至2016年4月于广州胸科医院住院治疗的42例肺结核患者作为肺结核(TB)组,健康对照(HC)组40名来自中山大学附属第一医院门诊体检的健康志愿者。收集研究对象血浆,分离出PBMC,采用流式细胞术检测各组PBMC和T淋巴细胞上OX40的表达;检测Th1、Th2、Th9和Treg细胞的比例。结果肺结核组PBMC、CD4~+T、Treg细胞上OX40的表达以及OX40~+细胞比例均明显低于健康对照组,差异有统计学意义(P均<0.05);肺结核组CD4~+T细胞中Th1、Th2和Treg亚群比例均明显高于健康对照组,而Th9亚群比例明显低于健康对照组,差异均有统计学意义(P<0.05);Treg细胞比例和Treg细胞上OX40~+比例呈负相关(r~2=0.347 4,P<0.05)。结论肺结核患者外周血CD4~+T淋巴细胞OX40的表达明显降低,并且肺结核患者Treg比例的增加可能与其OX40表达降低有关。
【Abstract】 Objective To compare the OX40 expression level in peripheral blood mononuclear cell(PBMC) and Tlymphocytes,as well as the percentage of T-helper cell subsets including Th1,Th2,Th9 and Treg,in pulmonarytuberculosis patients and healthy-control people. To further investigate the correlation between OX40 expression andtuberculosis as well as the differentiation of CD4~+T cell subsets. Methods A total of 42 pulmonary tuberculosis(TB)patients were selected from Guangzhou Chest Hospital between November 2015 and April 2016. And 40 healthy people wereselected as healthy control(HC). After PBMCs were isolated from these donors by lymphocyte isolation,the expression ofOX40 and the ratio of Th1,Th2,Th9 and Treg in CD4~+T cells were measured by flow cytometry. Results The expressionof OX40 in PBMCs,CD4~+T cells and Treg in pulmonary tuberculosis group were significantly lower than those in healthycontrol group(all P<0.05). The ratio of Th1,Th2 and Treg in CD4~+T cells in pulmonary tuberculosis group weresignificantly higher than those in healthy control group,whereas the percentage of Th9 in pulmonary tuberculosis group wassignificantly lower than those in healthy control group(P<0.05). The ratio of Treg in CD4~+T cells was negatively correlatedwith the ratio of OX40~+in Treg(r~2=0.347 4,P<0.05). Conclusions The expression of OX40 in CD4~+T lymphocytes inpulmonary tuberculosis patients is significantly lower than those in healthy people. The increased Treg in pulmonarytuberculosis patients may due to the decreased OX40 expression.
