载黄芩苷聚乙烯吡咯烷酮纳米胶束的制备

【摘要】 采用薄膜分散法制备载黄芩苷的大豆卵磷脂-十二烷基硫酸钠-聚乙烯吡咯烷酮K30三元混合物纳米胶束,研究其制备工艺.利用超滤离心法分离游离黄芩苷,在紫外分光光度下测定胶束中黄芩苷的含量并计算其包封率.通过单因素实验考察黄芩苷与大豆卵磷脂比例、聚乙烯吡咯烷酮K30投量、十二烷基硫酸钠投量、黄芩苷投量以及水浴温度等因素对黄芩苷纳米胶束包封率的影响,筛选出影响显著的因素.采用L9(34)正交实验设计进一步确定最优处方条件:黄芩苷与大豆卵磷脂比例为1∶40,十二烷基硫酸钠投量为0.20g,聚乙烯吡咯烷酮投量为0.20g,黄芩苷投量为3.0mg.该条件下,黄芩苷纳米胶束的包封率可达(92.62±1.77)%.激光粒度分析仪结果显示,黄芩苷纳米胶束平均粒径为(188.1±3.37)nm,PDI为0.163,粒径分布较为均匀.透射电镜结果显示,黄芩苷纳米胶束的形态多呈圆形或类圆形,大小与激光粒度分析所得粒径范围基本吻合.

【Abstract】 Preparation of baicalin nano-micelle was studied,soybean lecithin-sodium dodecyl sulfatepolyvinylpyrrolidone K30 ternary mixtures were prepared by film dispersion method,separating the free drugs by ultracentrifugation and determing the encapsulation rate by UV spectrophotometry.Based on the results from single factor tests,orthogonal experimental design method was used to investigate the factors influence on envelopment rate of liposome.In this experiment,the optimum conditions for the preparation of nano-micelles were as follows:the ratio of baicalin and soybean lecithin 1∶40,sodium dodecyl sulfate dosage 0.20 g,polyvinyl pyrrolidone dosage 0.20 g,baicalin dosage of 3.0 mg.The mean diameter was(188.1±3.37)nm,PDI 0.163,and encapsulation rate(92.62±1.77)%.The method could be used to prepare baicalin nano-micelle of soybean lecithin-sodium dodecyl sulfate-polyvinylpyrrolidone K30.
