六堡茶化学成分对胰α-淀粉酶的抑制作用

【摘要】 采用3,5-二硝基水杨酸(DNS)比色法观察六堡茶成分体外对胰α-淀粉酶催化水解作用的影响,根据紫外光谱、荧光光谱特征描述六堡茶成分与胰α-淀粉酶相互作用并推测其作用机制。研究发现:六堡茶成分能够影响胰α-淀粉酶催化活性,呈现抑制特征,其IC50为1. 72mg/m L,非竞争性可逆抑制,Ki为2. 52mg/m L。紫外、荧光光谱表明:六堡茶成分与胰α-淀粉酶作用诱导紫外吸收蓝移,使荧光猝灭。

【Abstract】 In this study,3,5-2 nitro salicylic acid(DNS) colorimetric method was used to evaluate inhibitory potential of L iubao Tea constituents against alpha-amylase in vitro.The interaction characteristics between alpha-amylase and Liubao tea constituents were studied by using ultraviolet spectroscopy and fluorescence spectroscopy.The results showed that Liubao tea constituents inhibit alpha-amylase activity significantly in vitro with the IC50 as 1.72 mg/m L and Ki as 2.52 mg/m L.The inhibitory type is reversible via non-competitive inhibition.Spectroscopies suggest that alpha-amylase could induce the peak location red-shift in the UV spectra and fluorescence-quenching in the fluorescence spectra.
